Research for the Mechanism of Inhibition of Viral Propagation Through AIFM1 Interaction with Pseudorabies Virus UL16 Protein
Apoptosis-inducing factor 1(AIFM1)is a complex mitochondrial protein,which can optimize the function of mitochondrial respiratory chain and participate in redox metabolism.It is the first caspase-independent apoptotic protein discovered so far.In this study,AIFM1 was found to interact with pseudorabies virus(PRV)pUL16.The results of overexpression and RNA interference showed that AIFM1 inhibited the proliferation of PRV and pUL16 weakened this inhibition.The laser confocal and flow test showed that AIFM1 translocated from mitochondria to nuclei when PRV infected cells and AIFM1 enhanced apoptosis induced by PRV.The interaction mechanism between PRV pUL16 and host protein AIFM1 was investigated and the roles of these two proteins in viral replication,packaging and interaction with host were partially analyzed in the present study,which laid a foundation for further elucidating PRV infection mechanism.
