Preliminary Study on Isolation and Molecular Epidemiology of Duck Circovirus in 2020 in Jiangsu Province
In order to study the molecular characteristics and propagation method of duck circovirus(DuCV)in Jiangsu province,23 organs and tissues of ducks suspected of circovirus disease were collected in 2020 and ground for DNA extraction.The DuCV specific fragments was amplified by PCR and sequenced for genetic analysis.As a result,10 out of 23 samples were positive in PCR test and were used for the following experiments.The supernatant samples of PCR positive materials were inoculated into 9-day-old SPF duck embryos through chorioallasic membranes.The chorioallantoic membranes and allallic liquids were collected and then tested by PCR.Then,PCR positive materials were continued to be passaged.The results showed that DuCV was detected by PCR in most samples at the first passage and some samples became positive in allantoic membranes at the second and third passages but disappeared at the fourth passage.The samples from allantoic membranes showed higher detection rate than that of allantoic fluid.Genomic sequencing found that all the DuCV samples were classified as type 1.This study would be beneficial to research of the molecular biological characteristics of DuCV and provide a reference for the prevention and treatment of the disease.
