Species Identification and Subtype Analysis of Cryptosporidium Causing Calf Diarrhea on a Dairy Farm in Gansu
Neonatal calf diarrhea(NCD)severely affects calf health and causes huge economic losses to the farm.Cryptosporidiosis is one of the important causes of NCD.However,there have been few reports of NCD outbreaks caused by Cryptosporidium spp.In September 2022,calves under one-month old developed severe diarrhea even death on a dairy farm in Wuwei city of Gansu province.In order to find out whether the calf diarrhea was caused by Cryptosporidium,20 fecal samples were collected from diarrhea and non-diarrhea calves within one-month old examined by microscopy and nested PCR in this study.The results showed that 10 Cryptosporidium positive samples were detected by microscopic examination.Additionally,14(70%)samples were tested positive by PCR,including 8 diarrhea samples and 6 non-diarrhea samples.All PCR positive samples were sequenced and identified as Cryptosporidium parvum.These C.parvum positive samples were amplified with gp60 gene for subtype analysis of which 13 samples were sequenced successfully and all of them were ⅡdA19G1 subtype.This study discovered an outbreak of calf diarrhea caused by Cryptosporidium for the first time in Gansu province.The results were of great significance for in-depth understanding calf diarrhea in this area and would help develop precise strategies for preventing and controlling calf diarrhea.
