柔嫩艾美耳球虫棒状体颈部蛋白EtRON2L2特性与功能初步分析

利用RT-PCR、间接免疫荧光实验、免疫印迹实验、抗体中和实验以及BiFC等方法对柔嫩艾美耳球虫棒状体颈部蛋白2同源分子—EtRON2L2的特性和功能进行研究。结果发现EtRON2L2氨基酸序列与常规的EtRON2仅有26。63%的同源性,mRNA水平在第二代裂殖子中最高,而蛋白水平在第二代裂殖子和子孢子阶段最高;EtRON2L2在游离的第二代裂殖子和子孢子阶段主要分布于胞质,而在入侵后子孢子中主要分布于虫体表面;抗rEtRON2L2多克隆血清处理组明显抑制子孢子入侵细胞;EtRON2L2与EtAMA3之间存在互作关系。本研究为探究EtRON2L2在球虫入侵过程中的作用机制奠定基础。
Characteristics and Biological Function of Rhoptry Neck Protein EtRON2L2 of Eimeria tenella
The molecular characteristics and biological function of rhoptry neck protein RON2L2 of Eimeria tenella(EtRON2L2)were investigated in the present study using RT-PCR,indirect immunofluorescence assay,Western blot,antibody neutralization and BiFC.The results showed that the amino acid sequence of EtRON2L2 had 26.63%identity with EtRON2.The mRNA level of EtRON2L2 was the highest in the second generation merozoites while the translation level was higher in both the second generation merozoites and sporozoites.EtRON2L2 was mainly distributed in the cytoplasm of the free second generation merozoites and sporozoites while it was mainly present on the surface of the parasites after invading cells.The anti-rEtRON2L2 antibodies had a significant inhibitory effect on the invasion of sporozoites.At the same time,there was an interaction between EtRON2L2 and E.tenella apical membrane antigen 3(EtAMA3).The results of this study laid a foundation for further research on the function and mechanism of EtRON2L2 in the invading process of E.tenella.
