Characteristics and Biological Function of Rhoptry Neck Protein EtRON2L2 of Eimeria tenella
The molecular characteristics and biological function of rhoptry neck protein RON2L2 of Eimeria tenella(EtRON2L2)were investigated in the present study using RT-PCR,indirect immunofluorescence assay,Western blot,antibody neutralization and BiFC.The results showed that the amino acid sequence of EtRON2L2 had 26.63%identity with EtRON2.The mRNA level of EtRON2L2 was the highest in the second generation merozoites while the translation level was higher in both the second generation merozoites and sporozoites.EtRON2L2 was mainly distributed in the cytoplasm of the free second generation merozoites and sporozoites while it was mainly present on the surface of the parasites after invading cells.The anti-rEtRON2L2 antibodies had a significant inhibitory effect on the invasion of sporozoites.At the same time,there was an interaction between EtRON2L2 and E.tenella apical membrane antigen 3(EtAMA3).The results of this study laid a foundation for further research on the function and mechanism of EtRON2L2 in the invading process of E.tenella.
