Isolation,Identification and Pathogenicity of Porcine Epidemic Diarrhea Virus CH/HB/CZ01
In order to obtain local isolates of Porcine epidemic diarrhea virus(PEDV)for preliminary understanding of their biological characteristics and pathogenicity,virus isolation and identification were performed in in Hebei province.The isolated PEDV was analyzed for its genetic evolution and pathogenicity in piglets.The samples collected in this area were inoculated onto Vero cells for continuous blind passages and the PEDV CH/HB/CZ01 strain was isolated by RT-PCR detection and gene sequencing identification.The genetic evolution analysis of the S gene of this strain was carried out using the sequences of the PEDV references published in GenBank.The results showed that the sequence homology between this strain and the popular strains found in recent years in China was 97.1%-98.3%,which was significantly higher than that of CV777,LZC,DR13 and other traditional strains and thus this strain belonged to the GⅡb subgroup.The isolated C H/HB/CZ01 virus culture was inoculated into healthy piglets with 2.0 mL each.,The inoculated piglets developed clinical signs such as diarrhea and their rectal swabs were tested PEDV positive by RT-PCR at 2 days post inoculation.The results showed that the isolated CH/HB/CZ01 virus was a mutant with strong pathogenicity,which laid a preliminary foundation for the study of the genetic and biological characteristics of the new PEDV strains in Hebei province.
