金黄色葡萄球菌336型多糖偶联抗原的制备及免疫原性

采用溶葡萄球菌酶消化菌体释放金黄色葡萄球菌(Staphylococcus aureus)336型多糖(Type 336polysaccharides,336PS),梯度乙醇浓度浸提和用不同酶消化进行粗提,通过凝胶过滤层析纯化得到了336PS多糖,并对其纯度进行检测.将336PS通过ADH间桥法与BSA连接制备偶联抗原,采用凝胶过滤层析纯化,200~400 nm波长扫描鉴定,从层析波峰和检定波长判定偶联成功.加佐剂制备成疫苗免疫小鼠,进行抗体检测和小鼠免疫保护试验.结果表明,纯化的336PS纯度为685.1 g/kg;偶联抗原波峰较BSA前移,其原最高吸收波峰为212 nm,从206 nm向280 nm偏移.偶联抗原336PS BSA可以刺激小鼠产生抗336PS的免疫应答反应,并能够对免疫小鼠提供免疫保护作用.
The Staphylococcus aureus was digested by lysostaphin to release type 336 polysaccharides (336PS),gradient concentration ethanol and different enzymes digestion were used for crude extraction,and further purification by gel filtration chromatography was done to obtain pure 336PS.The 336PS were conjugated to BSA by ADH spacing method to prepare a conjugated antigen.And it was purified by gel filtration chromatography and identified by wave scanning from 200 nm to 400 nm.336PS-BSA conjugated antigens subunit vaccine which mixed with oil-adjuvant was used to immunize mice,then antibody detection and immune protection tests were carried out.The results showed that the purity of 336PS was 685.1 g/kg,the peak of the conjugated antigen was ahead of BSA and the highest absorption peak was 212 nm,which moved from 206 nm to 280 nm.336PS-BSA could elicit anti-336PS immune responses,and it could also afford protection for mice challenged by a homeotype strain.
