云南库蠓源蓝舌病毒血清5型的分离与鉴定

摘要
为了解云南省西双版纳州库蠓中蓝舌病毒(BTV)的流行情况,于2022年7~8月在西双版纳勐海和景洪采集库蠓,进行了蓝舌病毒的分离和鉴定.本研究采用qRT-PCR检测库蠓中的蓝舌病毒,127份检测样品中,BTV阳性样品为3份,阳性率2.36%.阳性样品接种到C6/36细胞连续传代,第三代分离培养物接种的细胞均出现明显病变.分离培养物经qRT-PCR鉴定含有BTV,基于L2基因序列分析表明分离毒株均为BTV-5型.三株分离株L2基因序列与BTV-5参考毒株(AJ585126.1)的相似性超过93%,与云南师宗BTV-5(KT945045.1)的相似性超过98%;L3基因序列与云南芒市BTV-4(JX560415.1)的相似性大于96.8%.结果表明,云南西双版纳地区库蠓中存在BTV-5型流行.本研究为蓝舌病毒传播媒介的研究以及开展蓝舌病的预防控制工作提供了数据资料.
Abstract
To investigate the prevalence of bluetongue virus(BTV)in Culicoides from Xishuangbanna,Yun-nan province,Culicoides were collected from Menghai and Jinghong in Xishuangbanna from July to August 2022,and bluetongue virus was isolated and identified.Bluetongue virus in Culicoides was detected by qRT-PCR.Among 127 test samples,3 were BTV positive,resulting in a positive rate of 2.36%.These posi-tive samples were inoculated into C6/36 cells for continuous passage,and the third-generation isolat-ed cultures exhibited a significant cytopathic effect(CPE).The isolated cultures were confirmed to con-tain BTV by qRT-PCR,and the isolates were all BTV-5 based on the sequence analysis of L2 gene.These L2 gene sequences of the 3 isolates shared more than 93%similarity with the reference BTV-5 strain(AJ585126.1)and over 98%similarity with the BTV-5 strain(KT945045.1)isolated from Shizong,Yunnan province.The simi-larity of the L3 gene sequence from the 3 isolates with BTV-4(JX560415.1)isolated from Mangshi of Yunnan province was more than 96.8%.These results underscore the prevalence of BTV-5 in Culicoides in Xishuangbanna,Yunnan province.This study provided data for the study of bluetongue virus vector and the prevention and control of bluetongue disease.
