Preparation of monoclonal antibodies against yellow fever virus and identification of their biological characteristics
The aim was to prepare monoclonal antibodies(McAbs)against yellow fever virus(YFV)and analyze their biological characteristics,and then screen out McAbs with lethality,which provided laboratory basis for development of clinical detection methods and specific drugs of YFV.Using the brain of lactating mice subpassaged YFV,BALB/c mice of 6-8 weeks old were immunized by conventional methods,and 14 McAbs against YFV were screened and prepared by cell fusion,immunofluorescence detection and limited dilution clones.The subtypes of McAbs were identified by ELISA,and the results showed that 2 McAbs were IgG2b subtype,1 was IgM subtype,1 was IgG2a subtype,and the rest were IgG1 subtype.The specificity of McAbs was identified by immunofluorescence method,found that 12 McAbs did not cross-react with the detected virus strains.qRT-PCR method and immunofluorescence method were used to evaluate the neutralization effect of McAb cell supernatant and ascites on YFV,the results showed that 3 strains of McAb had better neutralization effect.In this study,12 McAbs of YFV with high specificity were obtained,among which 3 had good neutralization effect on YFV.
