黄热病毒单克隆抗体的制备及其生物学特性的鉴定

制备黄热病毒单克隆抗体并分析其生物学特性,进而筛选出具有杀伤力的中和抗体,为黄热病毒临床检测方法及特效药的研发提供实验室基础.使用乳鼠脑进行黄热病毒的传代扩增,通过常规方法对6~8周龄的BALB/c小鼠进行动物免疫,经细胞融合、免疫荧光检测及有限稀释克隆后,筛选制备黄热病毒单克隆抗体共14株.经ELISA鉴定单抗亚型,发现有2株单抗为IgG2b亚型,1株为IgM亚型,1株为IgG2a亚型,其余均为IgG1亚型;经免疫荧光法鉴定单抗特异性,发现有12株单抗与所检测的病毒株均无交叉反应;经qRT-PCR方法及免疫荧光法评估单抗细胞上清液及其腹水对黄热病毒的中和作用,发现有3株单抗的中和效果较佳.本研究获得了 12株特异性较高的单克隆抗体,其中有3株单抗对黄热病毒具有良好的中和作用.
Preparation of monoclonal antibodies against yellow fever virus and identification of their biological characteristics
The aim was to prepare monoclonal antibodies(McAbs)against yellow fever virus(YFV)and analyze their biological characteristics,and then screen out McAbs with lethality,which provided laboratory basis for development of clinical detection methods and specific drugs of YFV.Using the brain of lactating mice subpassaged YFV,BALB/c mice of 6-8 weeks old were immunized by conventional methods,and 14 McAbs against YFV were screened and prepared by cell fusion,immunofluorescence detection and limited dilution clones.The subtypes of McAbs were identified by ELISA,and the results showed that 2 McAbs were IgG2b subtype,1 was IgM subtype,1 was IgG2a subtype,and the rest were IgG1 subtype.The specificity of McAbs was identified by immunofluorescence method,found that 12 McAbs did not cross-react with the detected virus strains.qRT-PCR method and immunofluorescence method were used to evaluate the neutralization effect of McAb cell supernatant and ascites on YFV,the results showed that 3 strains of McAb had better neutralization effect.In this study,12 McAbs of YFV with high specificity were obtained,among which 3 had good neutralization effect on YFV.
