MEK1/2-ERK1/2通路参与硒在高皮质醇水平下对山羊子宫内膜炎症反应的抑制作用

为揭示硒在高皮质醇水平下对山羊子宫内膜炎症通路MEK1/2-ERK1/2的影响,采用空怀奶山羊建立子宫内膜炎模型,观察皮质醇和硒对山羊生理指标、子宫内膜组织病理学变化、子宫分泌物以及MEK1/2-ERK1/2通路的影响.结果显示,连续注射7 d皮质醇后,山羊血清皮质醇可维持较高水平;连续补饲酵母硒21 d可维持山羊血清较高的硒水平.灌注大肠杆菌后,山羊的呼吸频率、心率、体温和外周血白细胞数、子宫分泌物中多形核白细胞(PMN)数量以及MEK1/2-ERK1/2通路关键因子表达均升高,子宫组织病理损伤加重.注射皮质醇后,山羊生理指标、子宫分泌物中PMN数量均下降,子宫组织损伤加重;MEK1和MEK2基因表达下降(P<0.05),MEK1/2-ERK1/2通路关键蛋白表达水平下降(P<0.05).与皮质醇和大肠杆菌共处理组的山羊相比,日粮中补饲酵母硒的山羊的生理指标、子宫分泌物PMN数量进一步下降,子宫组织损伤减轻,MEK1/2-ERKl/2通路被抑制(P<0.05).结论:日粮中补饲酵母硒可减轻高皮质醇水平下大肠杆菌诱导的山羊子宫内膜的炎症反应,其调控机制可能与MEK1/2-ERK1/2通路有关.
MEK1/2-ERK1/2 pathway participates in the inhibition of inflammation of goat endometrium with high cortisol level
To investigate the effect of selenium on the MEK1/2-ERK1/2 pathway in goat endometrium under a high cortisol level,the uterine infection model of dairy goats was established by intrauterine infusion of Escherichia coli.Additionally,the animal received intramuscular injection of cortisol and dietary supplementation with yeast selenium.The changes in the physiological indicators,the endometrial histology,the uterine secretion,and the MEK1/2-ERK1/2 pathway were detected.As a result,the administration of cortisol for 7 consecutive days maintained a high serum cortisol level.The supplementation of selenium for 21 days stably elevated the serum selenium level.Goats received E.coli treatment presented increased respiratory rate,heart rate,body temperature,peripheral white blood cell counts,and polymorphonuclear neutrophil(PMN)amount in uterine secretions,the endometrial damage,and the activation of MEK1/2-ERK1/2 pathway.Cortisol treatment ameliorated the changes in the physiological indicators,inflammation markers,and tissue injury,and reduced the gene and protein expressions related to MEK1/2-ERK1/2 pathway(P<0.05).In goats received dietary supplementation with yeast selenium,there was a further inhibition in the physiological indicators,the PMN amount in uterine secret ions,the uterine tissue damage,and the MEK1/2-ERK1/2 activation(P<0.05).In conclusion,dietary supplementation with yeast selenium alleviated endometrial inflammation of goats under high cortisol levels,and its regulatory mechanism may be related to MEK1/2-ERK1/2 pathway.
