为了探究某规模化养殖场奶山羊隐性乳房炎致病菌的情况,采用体细胞计数法对奶山羊乳样进行隐性乳房炎检测,对阳性乳样中的细菌进行分离纯化、形态学鉴定、生化鉴定及分子生物学鉴定.通过动物实验验证分离株的致病性,利用PCR检测致病菌携带的毒力基因,采用纸片法对致病菌进行药敏试验.结果显示,该羊场隐性乳房炎的阳性率为 68.9%;从 40 份乳样中分离并鉴定出 14 株细菌,其中 8 株金黄色葡萄球菌分离株对小鼠有较强致病性,分离株KM4 对小鼠半数致死量为 1.8×108 CFU/mL.毒力基因检测结果显示,hla、hlb、clfA、sec、fnbpA和tsst-1 检出率均为 100%.药敏结果显示,分离株对青霉素和氨苄西林耐药率为 100%,对林可霉素耐药率为 12.5%,对头孢曲松和头孢他啶敏感率为 37.5%,对其他药物全部敏感.
Isolation and identification of Staphylococcus aureus for subclinical mastitis in dairy goats and detection of virulence genes
To investigate the situation of pathogenic bacteria caused by subclinical mastitis in dairy goats on a large-scale farm.In this experiment,somatic cell counting was used to detect subclinical mastitis in milk samples from dairy goats,and bacteria in positive milk samples were isolated and purified using selective media,identified morphologically,biochemically and molecularly.The main pathogenic bacteria were identified by animal pathogenicity test,PCR reaction was used to detect virulence genes carried by pathogenic bacteria,and paper disk method was used to test drug susceptibility of pathogenic bacteria.The results showed that the positivity rate of subclinical mastitis in dairy goat farms was 68.9%.14 strains of bacteria were obtained from 40 milk samples,8 of the Staphylococcus aureus isolates were highly pathogenic to mice,and the isolate KM4 had a LD50 of 1.8×108 CFU/mL in mice.The virulence gene test results showed that the detection rate of hla,hlb,clfA,sec,fnbpA and tsst-1 was 100%.The drug sensitivity results showed that the isolates were 100%resistant to penicillin and ampicillin and 12.5%resistant to lincomycin.37.5%sensitive to ceftriaxone and ceftazidime,and sensitive to all other drugs.
