Evaluation of the effect and mechanism of glycyrrhetinic acid against Toxoplasma gondii
Toxoplasmosis are mainly controlled by drugs,so screening for new anti-toxoplasmosis drugs is of great important.This study evaluated the anti-T.gondii effect and potential mechanism of glycyrrhetinic acid(GA)in vitro.The CC50 of GA was evaluated on HFF cells using MTT assay.The IC50 of GA were determined by plaque assay,and the influence of glycyrrhetinic acid on tachyzoite invasion and intracellular proliferation were evaluated.The mitochondrial membrane potential and reactive oxygen species(ROS)of T.gondii were detected by the JC-1 probe and automatic microplate reader,respectively.The results showed that CC50 of GA is 351.2 μmol/L,and IC50 of GA was 28.02 μmol/L.It can inhibit T.gondii intracellular proliferation at higher concentrations by decreasing T.gondii mitochondrial membrane potential level and increasing ROS production.In conclusion,GA has the properties of anti-T.gondii,and has potential as anti-Toxoplasma candidate drug,which is worthy of further study.
