邓氏巴贝斯虫血清学诊断抗原的筛选

为了鉴定用于邓氏巴贝斯虫检测的潜在抗原,本研究通过蛋白质组学筛选邓氏巴贝斯虫感染后释放到宿主血液中的分泌性抗原,结合生物信息学、分子生物学技术和血清学反应分析并验证其作为诊断抗原的潜力.结果表明,蛋白质组学共鉴定到34种分泌性抗原,生物信息学分析显示,其中8种蛋白在近源物种中没有同源蛋白,为邓氏巴贝斯虫物种特异性抗原,6种蛋白能在真核细胞中表达,2种蛋白(ExoAg-1200和ExoAg-2451)可与邓氏巴贝斯虫阳性血清发生特异性反应,而与健康金黄地鼠血清无反应.上述结果表明,本研究获得了两种邓氏巴贝斯虫物种特异性抗原,为今后建立其血清学检测方法提供了生物材料.
Screening of serological diagnostic antigens for Babesia duncani infection
In order to identify potential diagnostic antigens for detecting Babesia duncani infection,exoantigens(ExoAgs)secreted into host sera were screened by quantitative proteomics,and bioinformatics analysis,molecular biology techniques,and serological reactions were used to analyze and validate their potential as diagnostic antigens.The results showed that a total of 34 ExoAgs were identified using proteomics techniques.Bioinformatics analysis revealed that 8 ExoAgs did not have homologous proteins in Apicomplexan parasites,thus,these 8 ExoAgs were regarded as B.duncani species-specific antigens.Further analysis found that 6 ExoAgs were successfully expressed in eukaryotic cells,and 2 proteins(ExoAg-1200 and ExoAg-2451)could react specifically with B.duncani positive sera,but no reaction with healthy hamster sera.These results indicate that 2 B.duncani species-specific antigens were obtained in this study,and our research may provide biomaterials for the establishment of B.duncani serological detection methods in the future.
