摘要

本研究旨在评估CpG和polyI∶C对口蹄疫(FMD)病毒样颗粒(VLPs)疫苗的免疫增强效果.以纯化的O型FMD VLPs抗原为基础,使用VLPs-CpG和VLPs-polyI∶C刺激小鼠骨髓源性树突状细胞(BMDC),通过流式细胞术分析CD40和CD86在BMDC中的表达水平.进一步将VLPs、VLPs-CpG和VLPs-poly I∶C分别与ISA206乳化制备疫苗,并免疫豚鼠,通过检测特异性抗体、中和抗体、脾淋巴细胞增殖以及攻毒试验评估CpG和poly I∶C对VLPs疫苗免疫效果的影响.结果显示,VLPs-CpG组和VLPs-poly I∶C组的CD86表达水平高于VLPs组,ISA206佐剂联合poly I∶C、CpG均能提高攻毒保护率,而且ISA206联合CpG可以加速特异性抗体及中和抗体反应,提高脾淋巴细胞增殖水平.本研究结果表明,CpG和poly I∶C均能增强FMD VLPs疫苗的免疫效果,具有潜在的开发应用价值.

Abstract

This study was to evaluate the immune-enhancing effects of CpG and poly I∶C on the foot-and-mouth disease(FMD)virus-like particles(VLPs)vaccine.Using purified serotype 0 FMD VLPs as anti-gens,the mice bone marrow-derived dendritic cells(BMDC)were stimulated with VLPs-CpG and VLPs-poly I:C followed by the expression levels of CD40 and CD86 in BMDC was analyzed using flow cytometry.Fur-ther,VLPs,VLPs-CpG and VLPs-poly I∶C were emulsified with ISA206 to immunize guinea pigs.The improve-ment of CpG and poly I∶C on immune effect of the VLPs vaccine was evaluated by detecting the specific an-tibodies,neutralizing antibodies and splenic lymphocyte proliferation,as well as virus challenge.Re-sults showed that the expression levels of CD86 in VLPs-CpG and VLPs-poly I∶C were higher than VLPs.The adjuvant ISA206 combined with poly I∶C and CpG could improve the protection against virus.ISA206 com-bined with CpG accelerated the responses of specific antibodies and neutralizing antibodies,as well as enhanced the proliferation of splenic lymphocytes.This study demonstrates both CpG and poly I∶C has application potential to enhance the immune effect of FMD VLPs vaccine.
