Molecular taxonomic and categorization of Hyalomma scupense based on CO Ⅰ,ITS2 and 16S rDNA genes
Classification of Hyalomma scuppense is still unclear.In order to clarify the classifi-cation of Hyalomma scupense and analyze its phylogenetic relationships.In this study,ticks para-sitizing on animal surface were collected from Shache County,Xinjiang Uygur Autonomous Region,morpho-logical observation was carried out to identify the species of ticks.After extracting DNA samples from ticks,PCR,sequence analysis,and species identification were performed on the DNA samples based on the CO Ⅰ,ITS2,and 16S rDNA genes of ticks.A phylogenetic tree was constructed using MEGA 7.0.The results showed that the species of tick obtained in this study was preliminarily identified as Hyalomma scup-pense by morphological observation.The sequences of CO Ⅰ,ITS2,and 16S rDNA gene were compared with Hyalomma scuppense sequences that have been identified both domestically and internationally and ho-mologous analysis was performed.The sequence homology was 99.84%,99.52%and 98.42%,respectively,and the species of ticks obtained in this study was identified as Hyalomma Scutella.Phylogenetic trees showed that the CO Ⅰ,ITS2,and 16S rDNA sequences of the ticks obtained in this study were first clus-tered into a branch with the known Hyalomma scuppense sequences,and forming a large branch with the Hyalomma tick sequencs,and forming different branches from other tick species.The research results provide basic reference data for the distribution and identification of tick species in some areas of Xinjiang.
