Evaluation of the immune efficacy of STING agonist 22 immune enhancer combined with ASFV P30 protein in pigs
This study evaluated the immune effect of ASFV P30 target protein in pigs using STING ag-onist 22 as a candidate immune enhancer,and provided a preliminary experimental basis for screening the immunity booster of African swine fever(ASF)subunit vaccine.The P30 structural protein with high immunogenicity of prokaryotic expression of ASFV in our laboratory was selected as the target antigen and combined with STING agonist 22 immune enhancer and ISA 201VG adjuvant dosed with ASFV P30 protein for 2 immunization test pigs to compare the enhancement effect of STING agonist 22 immune enhancer in inducing cellular immunity,humoral immunity,and antigen delivery response.The results showed that the addition of STING agonist 22 immune enhancer group immunized test pigs significantly advanced the time of P30-mediated antibody response,increased the expression level of B cell maturation and differ-entiation-related transcription factors,and promoted the maturation of DCs and enhanced their antigen delivery,compared with 201VG adjuvant dosing alone.The results of this study showed that STING agonist 22 immune enhancer paired with ASFV P30 protein could significantly enhance cellular and humoral im-mune responses and induce stronger antigen presentation in pigs,and could be used as an immune enhancer for ASF subunit vaccines.
