摘要
为探究玉米赤霉烯酮(ZEA)对雄性小鼠铁死亡相关基因在睾丸组织中表达和定位的影响,以及β-葡聚糖对ZEA诱导小鼠睾丸细胞铁死亡的缓解作用.随机将50只雄性小鼠分为对照组(灌胃玉米油)、ZEA 组(LG:0.5 mg/kg、MG:5 mg/kg、HG:50 mg/kg)和β-葡聚糖组(TG:50 mg/kg ZEA+50 mg/kgβ-葡聚糖),连续灌胃10 d.用台盼蓝染色法、HE染色、免疫组化、Western-blot和qRT-PCR等方法检测小鼠精子和睾丸结构的变化及铁死亡基因GPX4、SLC7A11和FTH1的表达与定位.结果显示,与对照组相比,ZEA组畸形精子增多、曲精小管内生精细胞排列紊乱,睾丸组织Fe含量升高(P<0.01)、MDA含量升高(P<0.001)、SOD 含量下降(P<0.001),GPX4、SLC7A11 和 FTH1 的表达降低(P<0.05),GPX4 在精原细胞的细胞核表达,SLC7A11在精原细胞和精母细胞的细胞核表达,FTH1在精原细胞的细胞核和细胞膜表达.β-葡聚糖组与ZEA组相比畸形精子减少、生精细胞排列有序,Fe含量降低(P<0.05)、MDA含量降低(P<0.01)、SOD 含量上升(P<0.001),GPX4、SLC7A11 和 FTH1 的表达升高(P<0.05).结果表明,ZEA 可以通过降低GPX4、SLC7A11和FTH1在生精细胞的表达引起睾丸细胞铁死亡,酵母β-葡聚糖对ZEA诱导的小鼠睾丸细胞铁死亡有缓解作用.
Abstract
To investigate the effects of zearalenone(ZEA)on the expression and localization of ferroptosis-related genes in testicular tissues of male mice and the mitigating effects of β-glucan on ZEA-induced ferroptosis in mouse testicular cells.Fifty male mice were randomly divided into con-trol group(gavaged with corn oil),ZEA group(LG:0.5 mg/kg,MG:5 mg/kg,HG:50 mg/kg)and β-glucan group(TG:50 mg/kg ZEA+50 mg/kgβ-glucan)for 10 days.Taipan blue staining,HE staining,immunohistochem-istry,Western-blot and qRT-PCR were used to detect changes in the structure of mouse spermatozoa and testes and the expression and localization of the ferroptosis genes GPX4,SLC7A11 and FTH1.Compared with the control group,the ZEA group showed increased malformed spermatozoa,disorganized spermato-genic cells in the seminiferous tubules,elevated Fe content(P<0.01)and MDA content(P<0.001),de-creased SOD content(P<0.001),and decreased expression of GPX4,SLC7A11,and FTH1 in the testicular tissues(P<0.05).GPX4 was expressed in the nucleus of spermatogonia,SLC7A11 was expressed in the nu-cleus of spermatogonia and spermatocytes,and FTH1 was expressed in the nucleus and cell membrane of spermatogonia.Compared with ZEA group,β-glucan group had fewer abnormal sperm and more orderly sper-matogenic cells,Fe content(P<0.05),MDA content(P<0.01)decreased,SOD content increased(P<0.001),and the expression of GPX4,SLC7All,and FTH1 was elevated(P<0.05).The results showed that ZEA could cause testicular cell iron death by decreasing the expression of GPX4,SLC7A11 and FTH1 in spermato-genic cells,and that yeast β-glucan had an attenuating effect on ZEA-induced testicular cell ferrop-tosis in mice.
