Study on mechanism of host protein VDAC1 inhibiting replication of foot-and-mouth disease virus
foot-and-mouth disease is a highly contagious and severe illness triggered by the foot-and-mouth disease virus(FMDV)in even-toed ungulates like pigs,cattle,and sheep.The nonstructural protein 3Dpol of FMDV is crucial for the virus's replication and its role in causing the disease.In order to investigate the mechanism of host protein interaction with 3Dpol protein,this study firstly identi-fied the interaction between VDAC1 and FMDV 3Dpol using yeast two-hybrid and immunoprecipitation exper-iments,and then found the overexpression of VDAC1 and inhibitor experiments showed that VDAC1 inhibit-ed the expression of 3Dpol protein through autophagy pathway.Finally,it was found that down-regulation of VDAC1 had a promoting effect on FMDV replication,suggesting that VDAC1 was important in the replica-tion and pathogenicity of FMDV.This study reveals a new mechanism of host protein inhibition of FMDV replication,and the results provide a new direction and target for the prevention and control of FMD.
