摘要

为探究内蒙古地区牛病毒性腹泻的流行情况及牛病毒性腹泻病毒(BVDV)流行株基因特征,本研究采集了内蒙古某奶牛场腹泻犊牛的粪便样品,经过滤除菌等处理后,接种于牛肾细胞(MDBK),盲传5代后,观察细胞病变并进行RT-PCR鉴定.结果显示,成功分离到1株能致细胞病变的BVDV毒株,命名为BVDV-NM-1株;电镜观察到BVDV-NM-1株具有典型BVDV病毒粒子形态;对BVDV-NM-1株5'-UTR基因序列扩增后进行序列比对,结果显示,BVDV-NM-1株属于BVDV-1型,与BVDV-1v亚型的基因同源性为96%.本研究结果为内蒙古地区牛病毒性腹泻的防控奠定了基础.

Abstract

To investigate the prevalence of bovine viral diarrhea and the genetic characteristics of BVDV epidemic strains in Inner Mongolia area,In this study,fecal samples of diarrheal calves from a dairy farm in Inner Mongolia area were collected,filtered and sterilized,and inoculated into bovine kidney cells(MDBK).After 5 generations of blind transmission,the cytopathic changes were observed and identified by RT-PCR.The results showed that a cytopathic bovine viral diarrhea strain was successfully isolated and named BVDV-NM-1 strain.The typical BVDV virion morphology of BVDV-NM-1 was observed by electron microscopy.The 5'-UTR gene sequence of BVDV-NM-1 strain was amplified and compared with that of BVDV-1 type.The homology of NM-1 strain with BVDV-1v subtype was 96%.It laid the foundation for better prevention and control of bovine viral diarrhea in Inner Mongolia area.
