不同基因型弓形虫ROP16对人肺上皮细胞凋亡及细胞周期的影响

为探讨弓形虫Ⅰ型、Ⅱ型、Ⅲ型ROP16对人肺上皮细胞凋亡及细胞周期的影响及分子机制,本研究构建过表达Ⅰ、Ⅱ、Ⅲ型ROP16慢病毒载体并转染至人肺上皮细胞BEAS-2B细胞中,经嘌呤霉素筛选获得稳定过表达3种不同基因型ROP16细胞株,培养细胞并提取全蛋白及总RNA,通过RT-qPCR及Western-blot 检测 Ⅰ 型、Ⅱ 型、Ⅲ型 ROP16 对细胞凋亡相关蛋白 BAX、BCL-2、Caspase9(CAS9)、P53 及细胞周期相关蛋白CDK6、P21、Cyclin D1在mRNA和蛋白水平表达量的影响,并进一步探究不同基因型ROP16对细胞STAT3磷酸化水平的影响.Western-blot及RT-qPCR检测结果显示,3种不同基因型慢病毒过表达载体转染BEAS-2B细胞后其ROP16 mRNA和蛋白均明显表达,提示细胞模型构建成功;针对细胞凋亡及细胞周期相关蛋白的检测结果显示,与空白对照组及阴性对照组相比3种基因型ROP16均可引起细胞凋亡相关蛋白BAX和BCL-2下调,Ⅰ型和Ⅲ型ROP16可以引起CAS9及P53表达水平降低,且有统计学差异;Western-blot结果显示,Ⅰ型和Ⅲ型ROP16均可引起细胞周期相关蛋白P21、CyclinD1及CDK6表达下调,并有统计学差异;此外,Ⅰ型和Ⅲ型ROP16 过表达组BEAS-2B细胞中STAT3表达下调,而在Ⅱ型ROP16的BEAS-2B细胞中P-STAT3表达下调,且均有统计学意义.综上所述,Ⅰ型和Ⅲ型弓形虫ROP16相比于Ⅱ型可更显著的调节BEAS-2B细胞凋亡和细胞周期相关因子的表达及细胞STAT3磷酸化程度.
Effection of different genotypes of Toxoplasma gondii ROP16 on apoptosis and cell cycle of human lung epithelial cells
To investigate the effects and molecular mechanisms of Toxoplasma gondii type Ⅰ,Ⅱ,andⅢROP16 proteins on apoptosis and cell cycle in human lung epithelial BEAS-2B cells.Lentiviral over-expression vectors were constructed for overexpression of type Ⅰ,Ⅱ and Ⅲ ROP16 and transfected into BEAS-2B cells,which were screened by puromycin to obtain stable overexpression of the three different genotypes of ROP16 cell lines,and the cells were cultured and the whole proteins and total RNA were extracted.The effects of type Ⅰ,type Ⅱand typeⅢ ROP16 on the relative expression of apoptosis-asso-ciated proteins BAX,BCL-2,Caspase9(CAS9),P53 and cell cycle-associated proteins CDK6,P21 and Cyclin Dl at the Mrna and protein levels were detected by Western-blot and RT-Qpcr,and the effects of the ROP16 on the phosphorylation level of STAT3 in cells were also further investigated.Western-blot and RT-Qpcr results showed that three different genotypes of lentiviral overexpression vectors transfected BEAS-2B cells with ROP16 Mrna and protein were obviously expressed,suggesting that the cell model was successfully constructed.The detection results of apoptosis and cell cycle associated proteins showed that compared with the blank control group and negative control group,all three genotypes of ROP16 could cause down-regulation of apoptosis-related proteins BAX and BCL-2,and the expression levels of CAS9 and P53 could be reduced by type Ⅰ and type Ⅲ ROP16,and there was a statistically significant difference;Western-blot results showed that compared with the blank control group and negative control group,the expression levels of cellcycle-associated proteins P21 and Cyclin D1 were down-regulated by the three genotypes of ROP16,and CDK6 expression was down-regulated by the three genotypes.CDK6 expression was down-regulated in type Ⅰ and type Ⅲ ROP16 compared with the blank control and negative control groups,and the expression of STAT3 was down-regulated in BEAS-2B cells of type Ⅰ and type Ⅲ ROP16 com-pared with the blank control and negative control groups,while the expression of P-STAT3 was down-reg-ulated in BEAS-2B cells of type Ⅱ ROP16,and both of them were statistically significant.In conclu-sion,Type Ⅰ and Ⅲ Toxoplasma gondii ROP16 more significantly regulate the expression of apoptosis and cell cycle-related factors and cellular STAT3 phosphorylation in BEAS-2B cells compared to type Ⅱ.
