Objective To investigate the expression levels of GSDME and Caspase-3 in bone marrow CD34+cells of myelodysplastic syndrome(MDS)patients.Methods From September 2020 to December 2021,20 normal controls and 32 patients with myelodysplastic syndrome admitted to the hematology department of Tianjin Medical University General Hospital were compared,expression of GSDME and Caspase-3 in bone marrow CD34+cells was detected by Flow Cytometry and qRT-PCR.Results According to the Flow Cytometry,the expression levels of GSDME and Cleaved Caspase-3 of CD34+cells in bone marrow of low risk group were increased[(64.90±15.74)%,(31.83±17.99)%]than the control group[(41.39±14.07)%,(24.11±16.28)%,P<0.05];The expression of GSDME and Cleaved Caspase-3 in the high risk group[(30.69±14.62)%,(15.42±13.34)%,P<0.05]were decreased.According to the result of qRT-PCR,the relative expression of GSDME(0.20±0.19 vs.1.00±0.53,P<0.05)gene in high risk MDS patients was significantly decreased,but there was no significant difference in the expression of Caspase-3.Conclusion In low risk MDS patients,the expression of GSDME in CD34+cells increase,the level of pyroptosis may increase,leading to bone marrow hematopoietic failure.In high risk MDS patients,the expression of GSDME in CD34+cells decrease,the level of pyroptosis may decrease,the cells may proliferate excessively,and the risk of leukemia transformation is increased.
