Isolation,identification and genome sequencing of BPIV3 genotype B
To investigate the infection of bovine parainfluenza virus type 3 in herd of calves aged 1-6 months in an intensive cattle farm in Xinjiang,60 nasal swab samples were subjected to RT-PCR assay,virus isolation and identification,whole genome sequencing and genetic evolutionary analy-sis.The results showed that the nucleic acid positivity rate of BPIV3 was 11.67%.A strain of BPIV3 was isolated from the nucleic acid positive sample and named XJ21032-1(45),its genome length was 15 512 bp.Genetic evolutionary analysis showed that XJ21032-1(45)belonged to BPIV3 genotype B,and the highest homology was 93.4%with the Australian BPIV3 genotype B reference strain Q5592(EU277658).This study successfully isolated a strain of B-genotype BPIV3,confir-ming the existence and prevalence of B-genotype strains in China,providing raw materials for the development of BPIV3 vaccines,and contributing to further research on the molecular evolution and traceability of BPIV3.
