摘要

鸭短喙矮小综合征(short beak and dwarfism syndrome,SBDS)是近年来在我国鸭群中流行的一种新传染病,为研制SBDS灭活疫苗,本研究将SBDSV M15株在番鸭胚增殖,收获病毒尿囊液经灭活后制备了 3批灭活疫苗,并对疫苗的安全性、免疫原性及免疫效力进行检验.结果显示,3 日龄樱桃谷鸭超剂量接种1.0 mL/羽,未见不良反应,表明疫苗的安全性好;3批灭活疫苗免疫20日龄雏半番鸭,免疫后28 d血清的胶乳凝集抑制试验(LPAI)抗体效价均值为4.3 log2;130日龄樱桃谷鸭种鸭免疫1次灭活疫苗后60 d血清LPAI抗体平均效价为6.9 log2;种鸭免疫后60 d所产种蛋卵黄抗体的LPAI效价均值为6.5 log2;对种鸭免疫后的3日龄子代雏鸭进行攻毒获得100%免疫保护.结果表明,本研究研制的SBDS灭活疫苗具有良好的免疫原性,种鸭1次免疫后可提供子代雏鸭保护性母源抗体,保护雏鸭度过最易感的危险窗口期,为SBDS的防控提供了有效手段.

Abstract

Short beak and dwarfism syndrome(SBDS)is an infectious disease prevalent in Chinese duck flocks in recent years.In order to develop a SBDS inactivated vaccine,SBDSV M15 strain was inoculated in Muscovy duck embryos,and the allantoic fluid was harvested and inactivated by formaldehyde.Three batches of oil adjuvant inactivated vaccine were prepared,and the safety,im-munogenicity and immune efficacy of this vaccine was evaluated.The results showed that the vac-cine has good safety to 3-day-old Cherry Valley ducklings,no any adverse reactions were observed after injection an overdose of 1 mL per duck.Twenty-day-old mule duckling were immunized with three batches of inactivated vaccine and the mean sera antibody titer detected by latex particle ag-glutination inhibition assay(LPAI)was 4.3 log2 at 28 days post vaccination.High level of LPAI antibody was also induced in 130-day-old breed Cherry valley ducks with one dose vaccination,the average titers of LPAI antibody in sera and egg yolk were 6.9 log2 and 6.5 log2 at 60 d respectively.Complete immune protection was obtained when 3-day-old offspring ducks containing maternal antibodies were challenged with the pathogenic strain SBDSV M15.Taken together,the inactivated SBDS vaccine developed in this study has good immunogenicity,breed ducks with a single immuni-zation could offer offspring ducklings with high level of maternal antibodies to protect the duck-lings against SBDSV infection through the susceptibility period,providing an effective means for the prevention and control of SBDS.
