唾液乳杆菌CICC23174株全基因组测序及功能分析

为探究唾液乳杆菌CICC23174株的生物学特性和基因功能,本研究以从鸡肠道分离得到的唾液乳杆菌CICC23174株为研究对象,通过使用二代测序平台HiSeq2000进行测序,利用多种生物学信息软件对原始数据进行组装和优化,并对其基因信息进行功能注释、细菌素和信号肽预测、基因共线性比较和分子进化树分析.基因预测结果表明,唾液乳杆菌CICC23174株基因组大小为203 542 bp,GC含量约为32.84%,基因组特征显示其编码基因1 890个,含有3个细菌素和50个信号肽.重要的是,唾液乳杆菌CICC23174株含有27个特有基因,构成4个系统分别为Ⅰ型-CRISPR-Cas基因防御系统、Ⅱ型毒素-抗毒素系统、肿瘤抑制基因和蛋白分泌途径.而且基因共线性分析发现CICC23174株与模式菌株UCC118唾液乳杆菌最为相似,但是16S rRNA分子进化树结果表明CICC23174株与唾液乳杆菌FZJTZ9M6株遗传距离较近.本研究结果为丰富唾液乳杆菌的物种进化库,挖掘唾液乳杆菌潜在的益生功能奠定了基础.
Whole genome sequencing and function analysis of Ligilactobacillus salivarius CICC23174 strain
To study the biological characteristics and gene function of Ligilactobacillus salivarius CICC23174 strain,a strain was isolated from chicken intestines.The next generation sequencing platform HiSeq2000 was used for sequencing,and bioinformatics software was employed to assem-ble and optimize the raw data.Functional annotation,bacteriocin and signal peptide,gene collineari-ty comparison,and phylogenetic analysis were performed on its gene information.The results indi-cated that the linear genome of Ligilactobacillus salivarius CICC23174 strain was 203 542 bp in length,with a GC content of 32.84%.encoding 1 890 genes,containing 3 bacteriocins,and 50 signal peptides.Importantly,the Ligilactobacillus salivarius CICC23174 strain included 27 unique genes,forming four systems:the Type Ⅰ CRISPR-Cas defense system,the Type Ⅱ toxin antitoxin sys-tem,the tumor inhibitory gene,and the protein secretion pathway.Moreover,collinearity analysis showed that CICC23174 strain was most similar to the Ligilactobacillus salivarius UCC118 strain,but 16S rRNA phylogenetic analysis indicted that CICC23174 strain is genetically close to FZJTZ9M6 strain.This study completed the species evolution library of Ligilactobacillus salivari-us,and also lay the foundation for the investigation into the potential probiotic functions of Ligi-lactobacillus salivarius.
