Regulatory effect of chicoric acid on immunosuppression of macrophages induced by phosphoramide mustard
This study aims to establish a macrophage immunosuppression model in vitro and ex-plore the regulatory effect of chicoric acid(CA)on phosphamide mustard(PM)-induced immuno-suppression of RAW264.7 cells,providing a theoretical basis for the development of CA as a new veterinary immune enhancer.The cellular immunosuppression model was established by inducing with a concentration of 3 μmol/L PM for 24 h in RAW264.7 cells.Then cells were treated with dif-ferent concentrations of CA(25,50,100 μmol/L)for 24 h to investigate the regulatory effects of CA on immunosuppression.Cell viability was determined by CCK-8 method,cellular phagocytic ca-pacity was determined by neutral red phagocytosis method,IL-1β and IL-6 expression were determined by ELISA kit,NO content assay kit was used to measure NO level,and real-time PCR was used to determine the relative expression of IL-1β,IL-6 and iNOS mRNA.The results showed that macrophage viability and phagocytic capacity were significantly reduced(P<0.05)after RAW264.7 cells were induced by 3 μmol/L PM for 24 h,the expressions of NO,IL-1β,IL-6 and the corresponding relative expression of mRNA were decreased significantly(P<0.05),which re-vealed the macrophage immunosuppression model was successfully established.After treatment for 24 h with different concentrations of CA(25,50,100 μmol/L),macrophage viability and phagocyt-ic capacity were significantly enhanced(P<0.05),and the secretion levels of NO,1L-1β,IL-6 and the corresponding relative expression of mRNA were increased(P<0.05)in macrophages.To sum up,PM can effectively establish an immunosuppressive model of macrophages in vitro,CA may enhance the immune function of the body by improving the vitality and phagocytic ability of mac-rophages,promoting the expression of inflammatory factors,and exerting an immune enhancing effect.
