摘要
重组病毒载体是当前病毒基因工程研究的热点之一.羊口疮(orf),也称羊传染性脓疱皮炎,是由羊口疮病毒(orf virus,ORFV)引起的急性、接触性、嗜上皮性人畜共患传染病.ORFV为痘病毒科(Poxviridae)、脊索动物痘病毒亚科(Chordopoxrinae)的副痘病毒属(Parapoxvirus)成员,线性双股DNA病毒,病毒基因组较大,改造后可容纳较大的外源基因,适合开发为活病毒载体.本文综述了orf重组活病毒载体的优势、构建策略及其应用研究的最新进展,为ORFV研究、重组活病毒开发应用提供参考.
Abstract
Recombinant virus vector has become one of the current hotspots in viral genetic engi-neering.Orf,also known as contagious pustular dermatitis,is an acute,contact,epitheliophilic zoo-noses that is caused by orf virus(ORFV),the type species of the genus Parapoxvirus within the poxvirus subfamily Chordopoxvirinae.ORFV is linear double-stranded DNA(dsDNA)virus with a large viral genome that can accommodate large exogenous genes after transformation.It is suit-able for developing as a live viral vector.In this paper,the advantages,vector modification strate-gies,and application of recombinant live viral vector were reviewed,which can provide reference for the research of ORFV and the development and application of recombinant live virus.
