昆明地区某奶牛场致犊牛腹泻大肠杆菌的分离鉴定及生物学特性分析

为查明昆明地区某奶牛场致犊牛腹泻细菌性病原及其生物学特征,对腹泻犊牛粪便进行细菌分离培养、生化试验和16S rRNA序列分析以鉴定细菌种类,并对分离菌株进行致病性试验、血清型鉴定、毒力基因检测、药敏试验、耐药基因检测和种群分型鉴定.结果显示,从腹泻犊牛粪便中共分离鉴定4株强致病性的大肠杆菌,O3、O4、O103为优势血清型;4株致病性大肠杆菌对庆大霉素、头孢唑林、氯霉素、环丙沙星、诺氟沙星均表现强耐药性,而对丁胺卡拉表现为中度敏感;分离菌株携带blaOXA、blaTEM、aac(6')-Ib、oqxAB、aadA1共5种耐药基因及fimH、Crl、iu-cD、astA、hlyE、ompA、ompC、K88、papC等9种毒力基因,且4株致病菌均为B2型大肠杆菌.结果表明,导致昆明地区某奶牛场犊牛腹泻的病原为B2型致病性大肠杆菌,并对临床上常见的抗生素表现较强的耐药性,且携带多种耐药基因与毒力基因.本研究为该奶牛场犊牛腹泻选择有效的治疗药物提供了参考依据.
Isolation,identification and biological characteristics of Escherichia coli from di-arrhea of calves in a dairy farm in Kunming
To clarify the pathogenic bacteria causing calf diarrhea and the biological characteristics of pathogenic bacteria in a dairy farm in Kunming,the bacterial isolation and culture,biochemical tests and 16S rRNA assays were performed on calf diarrhea feces to identify bacterial species.In addition,pathogenicity tests,serotype,virulence gene,drug sensitivity,drug resistance gene and population typing detection were conducted on the isolated bacteria.The results showed that four strongly pathogenic Escherichia coli(E.coli)strains were isolated and identified from the feces of calves with O3,O4 and O103 as the dominant serotypes;the four pathogenic E.coli strains exhibi-ted strong resistance to gentamicin,cefazolin,chloramphenicol,ciprofloxacin and norfloxacin,while they were moderately susceptible to butylamine cara;the isolates had blaOXA,blaTEM,aac(6')-Ib,oqxAB,aadA1 and nine kinds of virulence genes such as fimH,Crl,iucD,astA,hlyE,ompA,ompC,K88,papC,and the four pathogenic strains were all type B2 E.coli.The results revealed that the type B2 pathogen causing calf diarrhea in a dairy farm in Kunming showed strong resist-ance to common clinical antibiotics and carried multiple resistance and virulence genes.This study provides a reference for the selection of effective therapeutic drugs for calf diarrhea in this dairy farm.
