Construction and biological characterization of oppCDF gene deficient strain of Salmonella typhimurium
The aim is to study the effect of oppCDF gene on the biological characteristics and path-ogenicity of Salmonella typhimurium(S.typhimurium).In this study,the oppCDF gene deletion strain of S.typhimurium was constructed by λ-Red homologous recombination technique,and its growth characteristics,motility,biofilm formation ability,intracellular survival ability and LD50 virulence were detected.The results showed that the growth characteristics of oppCDF gene dele-tion strains had no significant changes compared with the wild-type strains.oppCDF gene deletion can reduce the motility of S.typhimurium and affect the drug resistance of S.typhimurium in dif-ferent degrees.Deletion of oppC and oppF genes improved the biofilm formation ability of S.ty-phimurium.At the same time,oppC gene deletion reduced the survival ability in RAW267.4 cells and its virulence in mice,oppF gene deletion significantly enhanced the survival ability in RAW267.4 cells and its influence on the virulence of mice,while oppD gene deletion had no signif-icant changes in the virulence and survival rate in mice RAW267.4 cells.Studies have shown that oppCDF gene is closely related to the motility,biofilm formation and virulence of S.typhimurium.These results provide a theoretical basis for further revealing the complex regulation and mecha-nism of S.typhimurium pathogenicity.
