从下丘脑室旁核向背肩胛棕色组织发送投射的谷氨酸能神经元的鉴定

谷氨酸能神经元可抑制摄食并防止肥胖.有证据表明,在缺乏囊泡型谷氨酸转运蛋白2(VGlut2)的小鼠中,重新表达专一蛋白1(SIM1)神经元上的VGlut2可降低采食.然而,VGlut2神经元的位置和轴突投射尚不清楚.因此,本试验探寻了向背肩胛棕色脂肪组织(IBAT)发送神经元投射的上游脑区,检测了 VGlut2和黑皮质素4型受体(MC4R)神经元的脑内定位,确认了胰淀素(amylin)对于VGlut2和MC4R免疫阳性神经元表达的调控作用,拓展了VGlut2神经元发送神经支配的大脑区域.结果显示,PVNvGlut2神经元向IBAT发送密集的神经支配信号.在前皮质杏仁核(ACo)、纹状体(CPu)和海马齿状回(DG)发现 VGlut2与 MC4R共 定位.amylin注射显 著促进PVNvGlu2∶∶MC4R(P=0.004 8)免疫阳性神经元表达,提示VGlut2与MC4R涉及amylin的调节过程.PVNVGlut2神经元向下丘脑腹内侧核(VMH)、外侧下丘脑(LH)、中脑导水管周围灰质(PAG)和蓝斑(LC)核团发送神经元投射.鉴定了 VGlut2神经元及下游调节能量稳态的神经环路,为探究中枢神经系统和外周组织在能量稳态调节中可能发挥的联动作用提供证据.
Identification of glutamatergic neurons projecting from the hypothalamic paraven-tricular nucleus to interscapular brown adipose tissue
Glutamatergic neurons inhibit food intake and prevent obesity.There is evidence that re-expression of VGlut2 on specific protein 1(SIM1)neurons can reduce food intake in mice lacking vesicular glutamate transporter 2(VGlut2).However,the location and axonal projection of VGlut2 neurons are not clear.Therefore,in this study,we explored the upstream brain regions projecting to the interscapular brown adipose tissue(IBAT),detected the localization of VGlut2 and melanocor-tin type 4 receptor(MC4R)neurons in the brain,confirmed the regulatory effect of amylin on the expression of VGlut2 and MC4R neurons,and finally expanded the brain region where VGlut2 neurons sent innervation.The results showed that PVNVG1ut2 neurons sent dense innervation signals to IBAT.Co-localization of VGlut2 and MC4R was found in the anterior amygdaloid nucleus(ACo),Caudate putamen(CPu)and hippocampal dentate gyrus(DG).Amylin injection signifi-cantly increased the expression of PVNVGut2∶∶MC4R neurons(P=0.004 8)neurons,suggesting that VGlut2 and MC4R were involved in the regulation of amylin.PVNVGlut2 neurons project to the ven-tromedial hypothalamic nucleus(VMH),lateral hypothalamus(LH),periaqueductal gray(PAG)and locus ceruleus(LC).The identification of VGlut2 neurons and downstream neural circuits reg-ulating energy homeostasis provides some evidence for exploring the possible linkage role of cen-tral nervous system and peripheral tissue in energy homeostasis regulation.
