摘要

CXCL2基因可影响多种疾病并介导炎症和自身免疫的发生,但其在奶牛乳房炎中的作用鲜有报道.为初步揭示CXCL2基因在奶牛乳房炎中发挥的作用,本试验通过转染siRNA特异性序列构建CXCL2基因沉默模型,采用LTA刺激牛乳腺上皮细胞(MAC-T)构建奶牛乳房炎细胞模型,并通过CCK-8法检测CXCL2基因沉默对 MAC-T细胞活力的影响;EdU染色法检测CXCL2基因沉默对MAC-T细胞增殖水平的影响;流式细胞术检测CXCL2基因沉默对MAC-T细胞凋亡水平的影响.结果显示,CXCL2基因沉默可使炎性 MAC-T细胞活力和增殖能力增加,细胞凋亡数减少.CXCL2基因沉默可通过提高细胞活力、促进细胞增殖和抑制细胞凋亡来减轻LTA诱导的MAC-T细胞的炎症反应,但具体的分子机制仍有待进一步研究.

Abstract

CXCL2 gene can affect many diseases and mediate inflammation and autoimmunity,but its role in cow mastitis is rarely reported.In order to preliminarily reveal the role of CXCL2 gene in dairy mastitis,CXCL2 gene was silenced by transfecting siRNA specific sequence and a dairy mas-titis cell model was constructed by stimulating MAC-T cells with LTA.The effect of CXCL2 gene silencing on MAC-T cell viability was detected by CCK-8 assay.The proliferation of MAC-T cells after CXCL2 gene silencing was detected by EdU staining.Apoptosis levels were measured by flow cytometry in MAC-T cells with CXCL2 gene silencing.The results showed that CXCL2 gene si-lencing could increase the viability and proliferation capacity of inflammatory MAC-T cells and re-duce the number of apoptosis.CXCL2 gene silencing alleviates LTA-induced inflammatory re-sponse of MAC-T cells by enhancing cell viability,promoting cell proliferation and inhibiting apop-tosis,but the specific molecular mechanism remains to be further studied.
