高剂量酒糟摄入对山羊肾脏组织结构、P53和NF-κB通路的影响

摘要
近年来,我国反刍动物生产中高剂量饲喂酒糟现象较为常见,长期高剂量酒糟使用可使动物出现酒糟慢性中毒,严重制约了我国牛羊产业的健康发展.为探索高剂量酒糟摄入对山羊肾脏组织P53、NF-κB通路的影响,揭示其对山羊肾脏损伤的部分病理机制.参试山羊随机分为对照组、试验组,3只/组.对照组山羊按照《肉羊饲养标准》(NY/T816-2004)进行饲喂,试验组用酒糟代替日粮中75%蛋白质和能量成分的饲粮进行饲喂,试验期为90 d.90 d后采集山羊肾脏组织,用HE染色法观察肾脏组织病理变化,用蛋白免疫印迹法(Western blot)和实时荧光定量聚合酶链式反应(Real-time PCR,RT-PCR)检测山羊肾脏组织中NF-κB、P53通路中相关mRNA和蛋白表达量的变化.结果显示,试验组山羊肾脏肾小球肿大,皮质部毛细血管充血,间质毛细血管出血,肾小管上皮坏死;肾脏髓质部间质毛细血管淤血严重,管腔狭小,内有大量红细胞.与对照组相比,试验组山羊肾脏P53通路中CytC mRNA表达极显著上升(P<0.01),NF-κB通路中NEMO、MIP-2、PTGS2 mRNA表达极显著上升(P<0.01),PTGS2蛋白表达显著上升(P<0.05),p50、p65 mRNA表达显著上升(P<0.05).结果表明,高剂量的酒糟摄入,可造成山羊肾脏组织损伤,激活P53通路和NF-κB通路诱导山羊肾细胞发生凋亡和炎症反应.
Abstract
In recent years,the phenomenon of high-dose distiller's grains in the production of rumi-nants in China is relatively common,and the long-term use of high-dose distiller's grains can cause chronic poisoning of distiller's grains in animals,which seriously restricts the healthy development of China's cattle and sheep industry.This study aims to explore the effect of high-dose distiller's grains intake on p53 and NF-κB pathways in goat kidney tissue,and to reveal the pathological mechanisms of distiller's grains on goat kidney injury.The goats were randomly divided into the control group and the experimental group,with three goats in each group.The control group was fed according to the goat feeding standard recommended by"Feeding standard of meat-producing sheep and goats"(NY/T816-2004),and the experimental group was fed with distillers'grains in-stead of 75%protein and energy components in the diet.The experiment lasted for 90 d.After 90 d,kidney tissues were collected,and the pathological changes of the kidney tissues was observed by HE staining method.The changes of mRNA and protein expression in NF-κB and p53 pathways in goat kidney tissues was detected by Western blot and RT-PCR.The results showed that glomeruli of goats in the experimental group were enlarged,cortical capillary were congested,interstitial capillary were hemorrhagic,kidney tubular epithelial was necrotic,interstitial capillary in the me-dulla part of the kidney showed severe congestion,narrow lumen,and a large number of red blood cells,and compared with the control group,the expression of CytC mRNA in p53 pathway of kid-ney of experimental group was extremely significantly increased(P<0.01),NEMO,MIP-2 and PTGS2 mRNA expressions in NF-κB pathway were extremely significantly increased(P<0.01),and PTGS2 protein expression was significantly increased(P<0.05),p50 and p65 mRNA expres-sions were significantly increased(P<0.05).The results reveled that high dose distiller's grains ingestion could damage to goat kidney tissue,induce apoptosis and inflammation of goat kidney cells by activating p53 and NF-κB pathways.
