新疆地区犊牛冠状病毒的病原调查及S基因的遗传进化分析

采集了新疆8个地区15个牛场421份4月龄内犊牛的肛拭子,应用RT-PCR方法扩增牛冠状病毒(bovine coronavirus,BCoV)N基因和S基因,对S基因蛋白序列进行遗传进化分析.结果显示,BCoV个体核酸阳性率为19.91%(67/421),7~30日龄犊牛阳性率为33.59%(44/131),有临床腹泻症状犊牛的核酸阳性率为18.21%(57/313),伊犁地区最高34.88%(15/43);获得3条S基因序列,全长分别为4 092、4 092、4 093 bp,与参考序列核苷酸同源性为98.88%~99.46%;遗传进化分析显示3条S基因与国内四川毒株SWUN/NMG-13(MW711304.1)和BCOV-China/SWUN/LN4(MK095182.1)核苷酸同源性最近,为99.3%~99.4%,均属于GⅡ型;与国内外17株参考毒株相比,所测毒株存在23个氨基酸位点突变,均在I113V、D115A、S501P和T510S位点出现突变.结果表明,新疆地区1~4月龄犊牛普遍存在BCoV感染,且S蛋白存在部分突变.本研究揭示了新疆牛场犊牛BCoV不同月龄和地域流行分布及其S蛋白的变异特征,为BCoV防制提供了依据.
Pathogen investigation of calf coronavirus in Xinjiang and genetic evolution analy-sis of S gene
To understand the epidemic distribution and genetic variation of bovine coronavirus(BCoV)in calves in intensive farms in Xinjiang,421 anal swabs of calves within 4 months of age from 15 farms in 8 regions of Xinjiang were collected.The N gene and S gene of BCoV were ampli-fied by RT-PCR,and the genetic evolution of S gene protein sequence was analyzed.The results showed that the positive rate of BCoV individual nucleic acid was 19.91%(67/421),the positive rate of 7-30 days of age was 33.59%(44/131),the positive rate of clinical diarrhea calf nucleic acid was 18.21%(57/313),and the highest in Ili area was 34.88%(15/43).Three S gene sequences were obtained,with a total length of 4 092,4 092 and 4 093 bp,respectively.The nucleotide homol-ogy with the reference sequence was 98.88%-99.46%.Genetic evolution analysis showed that the three S genes had the closest nucleotide homology with the domestic Sichuan strain SWUN/NMG-13(MW711304.1)and BCoV-China/SWUN/LN4(MK095182.1),ranging from 99.3%to 99.4%,all belonging to G Ⅱ type.Compared with 17 reference strains at home and a-broad,there were 23 amino acid site mutations in the tested strains,all of which occurred at I1 13 V,D1 15 A,S501P and T510S sites.The results showed that BCoV infection was common in calves aged 1-4 months in Xinjiang,and there were some mutations in S protein.The study re-vealed the epidemic distribution of BCoV in different months and regions of calves in Xinjiang cat-tle farms and the variation characteristics of S protein,which provides a basis for BCoV prevention and control.
