Regulatory role of PGD2 on expression of cytokines and damage-related factors in bovine endometrial tissue of cows infected with Escherichia coli
The aim of this study is to investigate the effect of endogenous and exogenous prosta-glandin D2(PGD2)on the expression of cytokines(IL-1β,IL-6,TNF-α)and injury related factors(HMGB-1)in the endometrium of cows infected with Escherichia coli(E.coli).The results showed that compared with the blank control group,the gene expression of COX-2 and H-PGDS in the endometrium of cows infected with E.coli was significantly increased(P<0.00 1).The secre-tion of PGD2 also significantly increased(P<0.001).Endogenous PGD2 inhibitors(H-PGDS Inhib-itor 1,HQL-79)can significantly inhibit pro-inflammatory cytokines)in the endometrium of cows infected with E.coli(IL-1β,IL-6,TNF-α).The secretion and expression of injury related factors(HMGB-1)in mice reduced tissue damage.The exogenous addition of PGD2 can significantly pro-mote the secretion of pro-inflammatory cytokines in the endometrium of cows infected with E.co-li,and significantly increase the expression of HMGB-1 and the degree of tissue damage.The above results indicate that PGD2 can regulate the expression of pro-inflammatory cytokines and injury related factors in the process of E.coli infection of bovine endometrial tissue,thereby exerting a regulatory effect on tissue damage caused by bacterial infection.
