黑果枸杞花青素通过ERK1/2信号通路促进低氧诱导H9c2大鼠心肌细胞增殖

选用H9c2大鼠心肌细胞为试验对象,通过1%O2低氧诱导H9c2大鼠心肌细胞24 h,构建体外H9c2大鼠心肌细胞低氧模型,旨在探讨黑果枸杞花青素介导ERK1/2信号通路对低氧诱导H9c2大鼠心肌细胞增殖的影响.试验设常氧组、花青素组(50 mg/L)、低氧组(O2=1%)和低氧+花青素(50 mg/L)组.qRT-PCR法检测ERK1/2信号通路相关因子 ERK1/2、MKP3、c-Myc 的 mRNA 表达;Western blot 法检测 p-ERK1/2、ERK1/2、MKP3 和 c-Myc 蛋白表达.结果显示,与常氧组相比,低氧组显著下调ERK1/2的mRNA表达(P<0.05),极显著上调 MKP3的mRNA和蛋白表达(P<0.01),显著下调c-Myc的mRNA和蛋白表达(P<0.05),极显著下调p-ERK1/2/ERK1/2的蛋白表达(P<0.01);与低氧组相比,低氧+花青素组显著下调 MKP3的mRNA和蛋白表达(P<0.05),显著上调c-Myc的mRNA和蛋白表达(P<0.05),显著上调p-ERK1/2/ERK1/2的蛋白表达(P<0.05).结果表明,花青素可能通过下调MKP3表达、上调p-ERK1/2/ERK1/2蛋白表达、激活ERK1/2信号通路,进而上调增殖因子c-Myc的表达,促进低氧诱导H9c2大鼠心肌细胞的增殖.本试验为黑果枸杞花青素作为保护高海拔地区动物低氧适应性功能的饲料添加剂提供了理论依据.
Anthocyanin of Lycium barbarum promotes hypoxia-induced cardiomyocyte pro-liferation in H9c2 rats through ERK1/2 signaling pathway
H9c2 rat cardiomyocytes was selected as the experimental subjects,and was induced un-der 1%O2 hypoxia for 24 h to construct an in vitro H9c2 rat cardiomyocyte hypoxia model.The aim was to explore the effect of anthocyanin of Lycium barbarum mediated ERK1/2 signaling pathway on hypoxia induced proliferation of H9c2 rat cardiomyocyte.The experiment consisted of a normoxic group,an anthocyanin group(50 mg/L),a hypoxia group(O2=1%),and a hypoxia+an-thocyanin group(50 mg/L).The mRNA expression of ERK1/2 signaling pathway related factors ERK1/2,MKP3,and c-Myc was detected by qRT-PCR.Western blot was used to detect the expres-sion of p-ERK1/2,ERK1/2,MKP3,and c-Myc proteins.The results showed that compared with the normoxic group,the hypoxic group significantly downregulated the mRNA expression of ERK1/2(P<0.05),extremely significantly upregulated the mRNA and protein expression of MKP3(P<0.01),significantly downregulated the mRNA and protein expression of c-Myc(P<0.05),and extremely significantly downregulated the protein expression of p-ERK1/2/ERK1/2(P<0.01).Compared with the hypoxia group,the hypoxia+anthocyanin group significantly down-regulated the mRNA and protein expression of MKP3(P<0.05),significantly upregulated the mRNA and protein expression of c-Myc(P<0.05),and significantly upregulated the protein ex-pression of p-ERK1/2/ERK1/2(P<0.05).The results showed that anthocyanins may down-regu-late the expression of MKP3,up-regulate the expression of p-ERK1/2/ERK1/2 protein,activate the ERK1/2 signaling pathway,up-regulate the expression of proliferation factor c-Myc,and pro-mote hypoxia-induced proliferation of H9c2 rat cardiomyocytes.The results of the experiment pro-vide a theoretical basis for the use of anthocyanins in Lycium barbarum as a feed additive to pro-tect the hypoxic adaptability of animals in high-altitude areas.
