Anthocyanin of Lycium barbarum promotes hypoxia-induced cardiomyocyte pro-liferation in H9c2 rats through ERK1/2 signaling pathway
H9c2 rat cardiomyocytes was selected as the experimental subjects,and was induced un-der 1%O2 hypoxia for 24 h to construct an in vitro H9c2 rat cardiomyocyte hypoxia model.The aim was to explore the effect of anthocyanin of Lycium barbarum mediated ERK1/2 signaling pathway on hypoxia induced proliferation of H9c2 rat cardiomyocyte.The experiment consisted of a normoxic group,an anthocyanin group(50 mg/L),a hypoxia group(O2=1%),and a hypoxia+an-thocyanin group(50 mg/L).The mRNA expression of ERK1/2 signaling pathway related factors ERK1/2,MKP3,and c-Myc was detected by qRT-PCR.Western blot was used to detect the expres-sion of p-ERK1/2,ERK1/2,MKP3,and c-Myc proteins.The results showed that compared with the normoxic group,the hypoxic group significantly downregulated the mRNA expression of ERK1/2(P<0.05),extremely significantly upregulated the mRNA and protein expression of MKP3(P<0.01),significantly downregulated the mRNA and protein expression of c-Myc(P<0.05),and extremely significantly downregulated the protein expression of p-ERK1/2/ERK1/2(P<0.01).Compared with the hypoxia group,the hypoxia+anthocyanin group significantly down-regulated the mRNA and protein expression of MKP3(P<0.05),significantly upregulated the mRNA and protein expression of c-Myc(P<0.05),and significantly upregulated the protein ex-pression of p-ERK1/2/ERK1/2(P<0.05).The results showed that anthocyanins may down-regu-late the expression of MKP3,up-regulate the expression of p-ERK1/2/ERK1/2 protein,activate the ERK1/2 signaling pathway,up-regulate the expression of proliferation factor c-Myc,and pro-mote hypoxia-induced proliferation of H9c2 rat cardiomyocytes.The results of the experiment pro-vide a theoretical basis for the use of anthocyanins in Lycium barbarum as a feed additive to pro-tect the hypoxic adaptability of animals in high-altitude areas.
