4种藏药提取物与抗生素对产气荚膜梭菌生物被膜的影响

采用微量肉汤稀释法测定3种方法提取的4种藏药提取物及12种抗生素对(Clostridium perfringens,C.per-fringens)的最小抑菌浓度(minimum inhibitory concentration,MIC),棋盘法检测藏药提取物与抗生素的联合抑菌效果,结晶紫染色法测定20株牦牛源产气荚膜梭菌生物被膜(biofilm,BF)成膜能力,并检测藏药提取物与抗生素单用及联合使用对BF的抑制效果.结果显示:水提诃子对45.0%的C.perfringens的MIC≤0.80 g/L;波棱瓜子75%乙醇提取物及马兜铃甲醇提取物对20株C.perfringens的MIC均≤0.40 g/L;强力霉素和氨苄西林对19株C.per-fringens 的MIC≤0.25 mg/L,万古霉素的MIC均≤0.50 mg/L;诃子75%乙醇提取物与庆大霉素联用抑菌效果较好,对70.0%的C.perfringens产生协同作用;20株产气荚膜梭菌均具有BF成膜能力,55.0%的菌株呈强成膜能力;被检抗生素中多黏菌素B对BF的抑制作用最强,在1/4MIC处有强抑制效果;被检藏药提取物中马兜铃甲醇提取物与诃子75%乙醇提取物在1/2MIC处对BF具有强抑制效果;庆大霉素与诃子75%乙醇提取物联用及多黏菌素B与马兜铃甲醇提取物联用在两者均为1/4MIC时对BF仍有强抑制效果.结果表明,4种藏药提取物对C.perfringens具有抑菌效果,对其BF的形成具有抑制作用,与抗生素联用能增强抑制效果,需进一步对藏药抑菌机制进行研究.
Effects of four Tibetan medicinal extracts and antibiotics on biofilm of Clostridi-um perfringens
In order to investigate the inhibitory effects of extracts of Tibetan medicines and antibiot-ics on biofilm(BF)of Yak-derived Clostridium perfringens(C.perfringens),the minimum inhibi-tory concentration(MIC)of extracts of four Tibetan medicines extracted by three methods and twelve antibiotics against C.perfringens was determined by micro broth dilution method,and the combined inhibitory effects of extracts of Tibetan medicines and antibiotics were examined by u-sing checkerboard method.Crystalline violet staining was used to determine the film-forming abili-ty of 20 strains of C.perfringens BF and to test the inhibitory effects of Tibetan medicine extracts and antibiotics alone and in combination on BF.The results showed that the MIC of aqueous ex-tract of myrobalan was ≤0.80 g/L for 45.0%of C.perfringens strains;the MIC of 75%ethanol extract of pedunculate Herpetospermuan seed and methanol extract of Slender dutchmanspi pe fruit were ≤0.40 g/L for 20 strains of C.perfringens strains.The MICs of doxycycline and ampi-cillin against 19 strains of C.perfringens were ≤0.25 mg/L,and the MICs of vancomycin were both ≤0.50 mg/L.The 75%ethanol extract of myrobalan was more effective in bacteriostatic combination with gentamicin,producing a synergistic effect on 70.0%of C.perfringens.All20 strains of C.perfringens had BF film-forming ability,and 55.0%of the strains showed strong film-forming ability.Among the antibiotics examined,polymyxin B had the strongest inhibitory effect on BF,with a strong inhibitory effect at 1/4MIC.Among the examined Tibetan medicine ex-tracts,methanolic extract of Slender dutchmanspipe fruit and 75%ethanol extract of myrobalan showed strong inhibitory effect on BF at 1/2MIC.Gentamicin in combination with 75%ethanol ex-tract of myrobalan,and polymyxin B in combination with methanolic extract of Slender dutch-manspipe fruit remained strongly inhibitory to BF at 1/4MIC concentration in both.It shows that the extract of Tibetan medicine has inhibitory effects on C.perfringens and inhibits the formation of its BF,and the combination with antibiotics can enhance the inhibitory effect,which needs to be further researched on the mechanism of bacteriostatic inhibition of Tibetan medicine.
