为探究转录激活因子3(activating transcription factor 3,ATF3)在猪血凝性脑脊髓炎病毒(porcine hemaggluti-nating encephalomyelitis virus,PHEV)复制过程中的作用,本研究通过 Western blot、qRT-PCR和间接免疫荧光试验检测PHEV感染后ATF3表达水平及亚细胞定位变化,并在干扰ATF3后检测PHEV含量及相关炎性细胞因子变化.结果显示,PHEV感染后,ATF3的表达水平显著上调,同时ATF3从细胞浆转入细胞核中启动下游基因表达;而干扰ATF3后,细胞内炎性因子IFN-β,IFN-γ,IL-6,IL-1β转录水平下调,PHEV含量显著增加.结果表明,PHEV感染后上调ATF3可动态调节神经元中的炎性因子并对机体产生保护作用.
Expression and role of ATF3 in PHEV infection
In order to explore the function of activating transcription factor 3(ATF3)in the porcine hemagglutinating encephalomyelitis virus(PHEV)replication process,the current investigation was carried out using Western blot,qRT-PCR,and indirect immunofluorescence assays to identify changes in ATF3 expression level and subcellular localization following PHEV infection.Addition-ally,alterations in PHEV content and associated inflammatory cytokines were detected following ATF3 interference.According to the findings,ATF3 expression was dramatically increased follow-ing PHEV infection,and it translocated from the cytoplasm to the nucleus to trigger downstream gene expression.Interfering with ATF3 reduced the transcription levels of inflammatory factors IFN-β,IFN-γ,IL-6,and IL-1β,while increasing the PHEV content.The results indicated that up-regulation of ATF3 after PHEV infection could dynamically regulate inflammatory cytokines in neurons and had a protective effect on the body.
