摘要
非洲猪瘟(African swine fever,ASF)是由非洲猪瘟病毒(African swine fever virus,ASFV)感染猪所引起的一种急性、出血性、高度接触性传染病.自2018年ASF传入我国以来,已经流行了 5年多,病毒已经演化出了低毒力的基因Ⅱ型和基因Ⅰ型弱毒株,以及基因Ⅱ型和Ⅰ型重组毒株,这为该病的防制和疫苗研发带来了巨大挑战.ASF减毒活疫苗(live attenuated vaccines,LAVs)能够诱导良好的免疫保护,具备商品化疫苗的潜力.鉴定和研究毒力相关基因(virulence-associated genes,V AGs)对于LA Vs的研发至关重要,也是ASFV研究的热点和难点.因此,对已鉴定的ASFV VAGs进行全面系统的综述,总结VAGs编码蛋白的作用机制,同时展望如何全面鉴定ASFV VAGs及其编码蛋白的作用机制,以及如何利用已知的VAGs科学设计安全高效的LAVs,以期为ASFV的致病机制及LAVs研发提供参考.
Abstract
African swine fever(ASF)is an acute,hemorrhagic,highly contagious disease in pigs caused by African swine fever virus(ASFV).Since its introduction into China in 2018,ASF has un-dergone over five years of spread,resulting in the emergence of low virulent strains of genotypesⅡ and Ⅰ ASFV,as well as recombinant strains of genotypes Ⅰ and Ⅱ,which poses a serious chal-lenge to the prevention and control of the disease and the development of vaccines.ASF live attenu-ated vaccines(LAVs)have the potential to induce robust immune protection and hold promise as commercialized vaccines.Identification and functional investigation of virulence-associated genes(VAGs)are the basis of the development of LAVs and represent a focus and challenge in ASFV research.The review aims to provide a systematic and comprehensive review and analysis of the currently identified ASFV VAGs,and summarize the impact of the VAG-encoded proteins.Simul-taneously,the article offers a visionary outlook on comprehensive identification of ASFV VAGs and elucidation of the underlying mechanisms of their encoded proteins,and how to scientifically design safe and highly efficient LAVs utilizing the known VAGs.The goal is to provide valuable references for understanding the pathogenic mechanisms of ASFV and advancing the development of LAVs.
