乳腺纤维化对奶牛养殖业造成巨大的经济损失,本研究的目的是探究β-谷甾醇(BSS)对TGF-β1诱导的小鼠乳腺纤维化是否具有缓解作用.试验分为空白对照组、模型组、模型+药物处理组,以乳腺灌注TGF-β1的方式构建小鼠乳腺纤维化模型,待模型构建完24 h收集小鼠乳腺组织.应用HE染色和Masson染色评价乳腺组织病理结构的变化情况;应用荧光定量PCR方法检测乳腺组织中collagen Ⅰ、α-SMA、vimentin(VIM)等纤维化相关蛋白的mRNA水平变化;应用 Western blot和免疫荧光技术检测collagen Ⅱ、E-cadherin(E-cad)和α-SMA用于评价乳腺组织中纤维化蛋白表达情况.结果显示,BSS预处理后可以显著缓解TGF-β1诱导的小鼠乳腺病理损伤,显著抑制collagenⅠ、α-SMA、VIM mRNA水平的升高,显著抑制collagenⅡ、α-SMA蛋白水平的升高和E-cad蛋白水平的降低.BSS通过抑制纤维化表型指标的升高有效缓解纤维化病变的发生发展.
Effect of β-sitosterol on TGF-β1-induced mammary fibrosis in mice
This study aims to investigate the effect of β-sitosterol(BSS)on TGF-β1-induced mam-mary fibrosis in mice.The experiment was divided into the blank control group,model group,and model+drug group.Mouse mammary fibrosis model was constructed by infusing TGF-β1 into the mammary gland.After the model was constructed for 24 h,mouse mammary tissue was collected,and HE and Masson staining were used to evaluate the changes of mammary histopathology.mR-NA levels of fibrosis-related collagen Ⅰ,α-SMA and VIM in breast tissues were detected by fluo-rescence quantitative PCR.Western blot detection of collagen Ⅱ,E-cad and α-SMA was used to e-valuate fibrotic protein expression in mammary tissue.The results showed that in vivo experi-ments,BSS pretreatment could significantly alleviate the pathological damage of mammary gland induced by TGF-β1,and significantly inhibit the expression of collagen Ⅰ,α-SMA and VIM mR-NA.The increase of collagen Ⅱ,α-SMA protein levels and the decrease of E-cad protein levels was significantly inhibited.
