中药复方"四神丸加减"对感染白痢沙门菌雏鸡免疫功能和肠道菌群的影响

摘要
鸡白痢沙门菌病是危害养鸡行业最严重的疾病之一.抗生素的过度使用导致动物源性细菌产生耐药性.而中药复方替代抗生素已成为相关研究的重点和热点.中药复方"四神丸加减"(Sishen Wan Plus and Minus,SSJ)具有涩肠止泻、益气活血、清热利湿解毒等功能,但是SSJ对鸡白痢沙门菌病的影响尚缺乏研究.本研究旨在研究SSJ对感染白痢沙门菌雏鸡的免疫功能和肠道菌群的影响.将90只1日龄雏鸡均分为6组,空白对照组、鸡白痢沙门菌组、SSJ低剂量治疗组(0.4 g/kg)、SSJ中剂量治疗组(0.8 g/kg)、SSJ高剂量治疗组(1.6 g/kg)、抗生素组.通过16S rRNA测序和ELISA等技术探究SSJ对鸡白痢沙门菌病的影响.研究结果显示,SSJ能够显著降低由白痢沙门菌引起的雏鸡绒毛长度降低以及炎性因子水平的升高(P<0.05).此外,SSJ也能显著缓解由白痢沙门菌引起的雏鸡免疫球蛋白指数的显著降低(P<0.05).16S rRNA测序结果显示,高剂量SSJ能够显著增加菌群的多样性以及有益菌的丰度.以上结果证实,添加SSJ能够改善白痢沙门菌感染引起的雏鸡免疫功能减弱和肠道菌群多样性的减少.
Abstract
Salmonella pullorum is one of the most serious diseases affecting the chicken industry.The substitution of Chinese medicine compound for antibiotics has become the focus and hot spot of related research.The traditional Chinese medicine compound Sishen Wan Plus and Minus(SSJ)has the functions of inhibiting intestinal diarrhea,invigorating Qi and promoting blood circulation,clearing heat and dampness and detoxifying,but the effect of SSJ on chicken pullorosalmonellosis is still lacking.The aim of this study is to investigate the effects of SSJ on the immune function and intestinal flora of chickens infected with Salmonella pullorum.In this study,90 1-day-old chicks were divided into 6 groups:the blank control group,Salmonella pullorum group,SSJ low-dose treatment group(0.4 g/kg),SSJ medium-dose treatment group(0.8 g/kg),SSJ high-dose treat-ment group(1.6 g/kg)and gentamicin sulfate group.H&-E staining and ELISA were used to in-vestigate the effect of SSJ on chicken Salmonella pullorum disease.The results showed that the low,medium and high doses of SSJ could significantly reduce the decrease of villus length and the increase of inflammatory factors caused by Salmonella pullorum(P<0.05),and the high dose of SSJ had the best effect.In addition,the low,medium and high dose groups of SSJ could also signifi-cantly alleviate the decrease of immunoglobulin index caused by Salmonella pullorum(P<0.05).16S rRNA sequencing results showed that the high-dose group of SSJ could significantly increase the diversity of flora and the abundance of beneficial bacteria.These results confirm that SSJ sup-plementation can improve immune function and intestinal flora diversity of chicks induced by Sal-monella pullorum infection.
