The quality of pork is related to the development of porcine skeletal muscle and intra-muscular lipid deposition, and skeletal muscle satellite cells have the potential to differentiate into myogenic and adipocytes. In order to analyse the effect of proliferation and differentiation of skele-tal muscle satellite cells on skeletal muscle formation,in this study, skeletal muscle satellite cells were isolated and purified from the longissimus dorsi muscle of Landrace pigs and Bama minipigs, and the expression of Pax7 was detected by immunofluorescence for cell identification. The mRNA expression of cell cycle-related factors,myogenic-related factors and adipogenic factors in skeletal muscle satellite cells of two pigs was detected by RT-qPCR,and the proliferation of both cells was detected by CCK-8.The results showed that two kinds of porcine skeletal muscle satellite cells ex-hibited different proliferative ability and differentiation potential:Compared with Bama minipigs, the skeletal muscle satellite cells from Landrace pigs showed higher proliferative ability and higher myogenic differentiation potential and lower adipogenic differentiation potential. In summary, this study found that there are differences in the proliferation and differentiation ability of porcine skel-etal muscle satellite cells in different meat-quality pigs, which provides new theoretical data for e-lucidating the mechanism related to the differences in skeletal muscle formation in different meat-quality pigs.
