Expression and clinical significance of c-Myc and CDK12 in gastric carcinoma tissue
Objective To investigate the expression of c-Myc and cyclin-dependent kinase 12(CDK12)in gastric carcinoma tissue and their correlation with clinical features and prognosis.Methods 80 specimens of gastric carcinoma and para-carcinoma tissues were collected.The expression of c-Myc and CDK12 was detected by immunohistochemistry.The positive expressions of c-Myc and CDK12 in gastric carcinoma tissues and para-carcinoma tissues were compared,and the correlation between positive expressions of c-Myc and CDK12 in gastric para-carcinoma tissues and clinicopathological features of patients was analyzed,as well as the correlation between positive expressions of c-Myc and CDK12 in gastric carcinoma tissues and the prognosis of patients.Results The positive expression rates of c-Myc and CDK12 in gastric carcinoma tissues(77.5%,87.5%)were significantly higher than those in para-carcinoma tissues(13.8%,15.0%)(P<0.05).There was no statistically significant difference in the positive expression rates of c-Myc and CDK12 in gastric carcinoma tissues of patients with different age,gender and maximum tumor diameter(P>0.05).The positive expression rates of c-Myc and CDK12 in gastric carcinoma tissue of patients with middle-low differentiation,stage Ⅲ-Ⅳ,serosa invasion and lymph node metastasis were 88.0%,87.0%,88.9%,90.0%and 94.0%,92.6%,97.2%,100.0%,which were significantly higher than those of 60.0%,57.7%,68.2%,70.0%and 76.7%,76.9%,79.5%,80.0%in gastric carcinoma tissue of patients with high differentiation,stage Ⅰ-Ⅱ,no serosa invasion and no lymph node metastasis(P<0.05).Correlation analysis showed that the expression of c-Myc and CDK12 were positively correlated with each other(r=0.487,P=0.016<0.05).The 3-year survival rate of patients with positive expression of c-Myc and CDK12 was significantly lower than that of patients with negative expression of c-Myc and CDK12(P<0.05).Conclusion The expression of c-Myc and CDK12 is abnormally high in gastric carcinoma tissues,and the expression of c-Myc and CDK12 is positively correlated with the TNM stage,differentiation,depth of invasion and lymph node metastasis of gastric carcinoma,and have a significant impact on the prognosis of patients.It may be helpful to evaluate the prognosis of patients with gastric cancer by detecting their levels.
