Comparison of the HE staining effect of frozen rapid fixative and conventional fixative
Objective To compare the hematoxylin-eosin (HE) staining effect of frozen rapid fixative and conventional fixative. Methods Selected 20 intraoperative frozen rapid specimens,cut the tissue into 4 μm thickness and adhere to 2 adhesive slides,blew the electric blower for 15 s,immediately conducted tissue fixation and HE staining with 95% alcohol and rapid fixative (ethanol) in the same steps and the same time,and then sealed with neutral gum. After the rapid end of intraoperative freezing,the remaining tissue was melted,and a sufficient amount of 10% neutral buffered formalin (NBF) was added for subsequent routine sampling,production and HE staining. Routine sections were compared with intraoperative frozen rapid production for HE staining. Results Overall,95% alcohol made the tissue hematoxylin staining shallow,eosin staining deep,and the overall staining was red. Intercellular spaces became larger in individual tissues. The contrast of hematoxylin and eosin in the frozen rapid fixative were better,and could clearly present the nucleus structure. NBF allowed better fixed tissue and good conventional HE staining. But from a single tissue:(ⅰ) Breast sentinel lymph node:in rapid frozen,the staining effect of rapid fixative was better than 95% alcohol,the nucleus structure was clearer than the latter,no wrinkle between cells,and the color of HE staining is more vivid,which was more convenient for diagnostic physicians to read the film. (ⅱ) Thyroid:compared with the rapid fixative,the staining effect of 95% alcohol was not ideal,and the hematoxylin staining was shallow,the eosin staining was deep,and the overall staining was dark. There was a phenomenon of tissue contraction. The staining effect of NBF was thick and heavy,with high color contrast and clear gland structure. (ⅲ) Gallbladder:after fixation with 95% alcohol and rapid fixative,the nucleus and gland structure were clear,the contrast of hematoxylin and eosin staining was good,while after fixation with NBF,the conventional HE staining effect of gallbladder was not as fast as that of freezing. (ⅳ) Uterine fibroids:the hematoxylin staining of 95% alcohol was shallow,and eosin staining was deep. The hematoxylin staining of the rapid fixative was deeper than 95% alcohol,making it easier for doctors to observe the slices. NBF stained well.Conclusion The staining effect of rapid fixative is better than 95% alcohol and is better for tissue fixation. While NBF is preferred for fixation of conventional pathology specimens.
