摘要
目的 探究临床子宫内膜异位症组及正常的子宫内膜组两者之间的差异性基因,寻找子宫内膜异位症诊断的新型标志物.方法 收集临床卵巢型子宫内膜异位症患者的囊肿组织8例及正常对照的子宫内膜6例行转录组测序,通过差异基因火山图、KEGG信号通路、GO富集分析,寻找差异基因及差异通路.选取测序有差异性的MAPK通路蛋白通过免疫组化验证及PCR验证.结果 转录组测序结果提示子宫内膜异位症组相较于正常组MAPK信号通路蛋白在子宫内膜异位症组,免疫组化及PCR结果显示MAPK6、MAPK10、MAPKBP1、MAPK8IP2在子宫内膜异位症组蛋白水平及mRNA水平均增高.结论 MAPK信号通路蛋白可能在子宫内膜异位症发生发展中发挥着重要的作用,为子宫内膜异位症的诊治提供新靶点.
Abstract
Objective To explore the difference of genes between endometriosis group and normal endometrium group,and to find a new marker for the diagnosis of endometriosis.Methods 8 cases of ovarian endometriosis and 6 ca-ses of normal endometrium were collected in RNA-seq analysis.Differential genes and differential pathways were found by volcano map,KEGG signal pathway and GO enrichment analysis.MAPK pathway proteins with different sequencing were selected and verified by immunohistochemistry and qRT-PCR.Results the results of transcriptional sequencing showed that the expression of MAPK signal pathway protein in endometriosis group was higher than that in normal group.The results of immunohistochemistry and qRT-PCR showed that MAPK6,MAPK10,MAPKBP1 and MAPK8IP2 were higher in histone and mRNA levels of endometriosis.Conclusion MAPK signaling pathway protein may play an important role in the occurrence and development of endometriosis and provide a new target for the diagno-sis of endometriosis.
