Evaluation of Antibody Detection Method for Bovine Tuberculosis in Bulk Tank Milk
In order to assess whether the detection of antibodies in bulk tank milk can be used for monitoring and early warning of bovine tuberculosis,this study utilized enzyme-linked immunosorbent assay(ELISA)antibody detection method to analyze the correlation of bovine tuberculosis antibodies between milk samples and their corresponding blood samples from 85 dairy cows in three large-scale dairy farms in Hubei Province.By detecting bulk tank milk from 27 dairy farms and 2 998 individual milk samples,the correlation between the antibody levels in bulk tank milk and the prevalence of bovine tuberculosis was analyzed.A predictive model for bovine tuberculosis prevalence based on bulk tank milk antibody detection was established.To determine the detection capability of different potency positive milk samples in bulk tank milk,positive milk samples were diluted and mixed for detection,and differences in the empirical distribution functions of antibody levels among individual animals with similar bulk tank milk S/P values were analyzed.The results showed a high consistency(Kappa=0.862)and correlation(Pearson correlation coefficient r=0.976,P<0.01)between bovine tuberculosis antibody detection results in milk samples and blood samples.There was a moderate correlation between bulk tank milk antibody levels and the prevalence of bovine tuberculosis(Pearson correlation coefficient r=0.681,P<0.01).The predictive ability of the prevalence model y=e4.701 0+1.878 4ln(x)-0.01(R2=0.893 3)was 89.33%.When diluting positive milk samples of different titers,the positive detection rates after 2-fold dilution of weak-positive milk samples,16-fold dilution of moderate-positive milk samples,and 16-fold dilution of strong-positive milk samples were 16.67%,50.00%,and 100%,respectively.The results indicate that milk samples can be used for bovine tuberculosis antibody detection,and there is a correlation between bulk tank milk antibody levels and the prevalence of bovine tuberculosis.The prevalence of bovine tuberculosis can be predicted through the model.Bulk tank milk antibody detection can be used as an economical and rapid detection means for monitoring and early warning of bovine tuberculosis,with higher sensitivity for farms with higher prevalence or the presence of strongly positive individuals.
