Analysis of Key Genes for Prognosis of Head and Neck Squamous Cell Carcinoma Based on TCGA Database
Objective To explore the key genes for the prognosis of head and neck squamous cell carcinoma by using the existing TCGA database for the expression profile and survival data of head and neck squamous cell carcinoma.Methods Gene differential expression analysis was performed based on machine learning method and with statistical significance.The genes related to survival were preliminarily screened out by LASSO regression and elastic network regression,and the screening results were double-tested based on the GEPIA database and Wilcox test.The survival analysis of the tested genes was carried out by using the UALCAN database and COX proportional hazards regression model,and the relationship between key genes in prognosis and survival was explored.Results A total of 1,881 differential genes were obtained,and 18 genes screened by elastic network regression passed the double test(P<0.05).Six genes were significantly correlated with the survival rate of head and neck squamous cell carcinoma in the UALCAN database and the COX proportional hazards regression model,among which CDKN2A and DMRTA2 were positively correlated with the survival rate of patients.PLAU,STC2,SYT1 and TIMP4 were negatively correlated with the survival rate of patients.Conclusion CDKN2A,DMRTA2,PLAU,STC2,SYT1 and TIMP4 are key genes for the prognosis of head and neck squamous cell carcinoma.
