The effect and mechanism of Saponins of Radix Glycyrrhiza in mice with acute pancreatitis
[Objective] To explore the effect and mechanism of Saponins of Radix Glycyrrhiza in mice with acute pancreatitis.[Methods] Sixty Balb/c mice were randomized into control group,model group,low dose group and high dose group.Acute pancreatitis mice wereinduced by intraperitoneal injection of 100 μg/kg caerulein.Mice in low dose group and high dose group were treated with 15 mg/kg and 45 mg/kg Saponins of Radix Glycyrrhiza at 3 h before model construction.Mice in four group were sacrificed at 8,16 and 24 hours.Serum level of amylase (AMY) was determined by automatic biochemistry analyzer.TNF-α and IL-6 were measured by ELISA.Pancreatic histopathological changes were observed by HE staining.Expressions of HMGB1 in pancreas were detected by Western blotting.[Results] Compared with control group,Serum AMY,TNF-α and IL-6 level,pancreatic histopathological changes and expressions of hmgb1 in pancreas in model group with the prolongation of time were significantly enhanced(P <0.05).Compared with model group,serum AMY,TNF-α and IL-6 level and expressions of hmgbl in pancreas in low dose group and high dose group significantlywere reduced,and pancreatic histopathological changes in low dose group and high dose groupwere significantly improved (P <0.05).Compared with low dose group,the index above of all in high dose group were more significantly improved(P <0.05).[Conclusion] Saponins of Radix Glycyrrhiza can improve inflammation level and alleviate histopathological changes bydown-regulating HMGB1 protein in mice with acute pancreatitis.
