急性T淋巴细胞白血病中HOXB簇基因表达的临床特征及预后

摘要
目的 回顾性分析HOXB簇基因在儿童T-ALL中的表达模式及其与患儿临床特征及预后的关系,以期为儿童T-ALL的精准化、个体化诊疗提供依据.方法 收集2012年7月1日—2022年9月30日期间诊疗于苏州大学附属儿童医院的115例初诊T-ALL患儿血液标本,采用全转录组测序检测HOXB簇基因表达量,比较HOXB簇基因高表达组与低表达组患儿的临床特征及治疗反应.结果 115例患儿中,10个HOXB簇基因只有HOXB2-7有高表达.HOXB2、HOXB4、HOXB7基因低表达组D15骨髓幼稚细胞≥25％的患儿多于高表达组(P值均＜0.05).HOXB 簇基因表达程度不影响T患儿的预后.结论 儿童T-ALL中HOXB2、HOXB4、HOXB7基因低表达组早期治疗反应差,但对T-ALL预后没有影响.
Abstract
Objective This study retrospectively analyzed the expression pattern of HOXB gene clusters in pediatric T-ALL,and the association with the clinical features and patients'prognosis.We aimed to provide evidence for precision and personalized therapy for pediatric T-ALL.Methods The clinical data of 115 pediatric patients with T-ALL treated at the Children's Hospital of Soochow University between July 1,2012 and September 30,2022 were retrospectively analyzed.The expression of te HOXB genes was detected by transcriptome sequencing.The patients were categorized into high-HOXB and low-HOXB expression groups,and their clinical characteristics and treatment response were compared.Results Only six of the ten HOXB genes were highly expressed,including HOXB2-7.The number of children with day 15 bone marrow blasts cell≥25％in the HOXB2,4,7 low expression group was significantly higher than those in the high expression group.But the expression level of the HOXB genes was not significantly associated with the prognosis of T-ALL patients.Conclusions Of the ten HOXB genes,six genes(HOXB2-HOXB7)were highly expressed in patients with T-ALL.Patients with low HOXB2,4,7 gene expression had no effect for the prognosis.
