视神经萎缩1型线粒体融合蛋白在心血管疾病中的研究进展

摘要
心血管疾病严重威胁着人类健康,现已成为人类死亡的主要原因.能量代谢异常对心血管疾病的发生和发展起着关键作用,人体内90%的能量来源于线粒体,因此线粒体功能的稳定对于维持细胞正常形态与功能至关重要.既往研究发现,位于线粒体内膜上的视神经萎缩1型(OPA1)对线粒体功能的维持至关重要,越来越多的研究也表明,OPA1在动脉粥样硬化、心肌缺血再灌注损伤、心力衰竭、糖尿病心肌病等心血管疾病中发挥重要作用.本综述将概述有关OPA1蛋白功能,功能障碍以及与心血管疾病相关的临床表型,重点关注治疗OPA1突变引起的相关心血管疾病的当前治疗选择和未来前景.
Abstract
Cardiovascular disease is a serious threat to human health and has become a major cause of death worldwide.In the course of cardiovascular disease research,it has gradually been found that abnormal energy metabolism plays a key role in the occurrence and development of the disease.Mitochondria produces 90%of the ATP in the human body,therefore the stability of mitochondrial function is essential for maintaining normal cellular form and function.Previous studies have found that optic atrophy type 1(OPA1),located on the inner mitochondrial membrane,is essential for the maintenance of mitochondrial function and has a protective effect against cardiovascular-related diseases such as atherosclerosis,ischemia-reperfusion injury,heart failure and diabetic cardiomyopathy.In this review,The paper outlines recent findings regarding OPA1 protein function,dysfunction and associated clinical phenotypes,with a focus on current therapeutic options and future prospects for the treatment of OPA1 mutation-induced related cardiovascular diseases.
