Objective To investigate the expression level of serum amyloid P component(SAP)in aorta of patients with aortic dissection and its potential clinical significance.Methods A total of 60 aortae samples were collected from patients with Stanford type A aortic dissection(TAAD)and 30 normal aortae samples were collected from heart transplant donors from January 2020 to December 2021 in the Division of Cardiovascular Surgery,Tongji Hospital,Tongji Medical College,Huazhong University of Science and Technology.The differences in the aortic wall morphology between the two groups were analyzed by hematoxylin-eosin(HE)staining,and elastic fiber(EVG)staining.The subcellular localization and the expression level of SAP in the aortic wall tissues were further examined by Western blotting,immunofluorescence staining and immunohistochemistry.Results HE and EVG staining showed that the aortic smooth muscle cells were disorganized and elastic fibers were broken and reduced in the aorta of patients with TAAD compared with the control group.The results of Western blotting,immunofluorescence staining and immunohistochemistry staining showed that the expression level of SAP protein was significantly reduced in the aortic wall tissue of patients with TAAD compared with the control group(P<0.05),and the results of immunofluorescence staining showed that SAP protein was mainly localized in the cytoplasm and extracellular matrix.Conclusions SAP protein reduces in the aortic wall tissues of aortic dissection patients,suggesting that SAP protein may be associated with the development of aortic dissection.
