补体C1q在动脉粥样硬化中的作用

摘要
动脉粥样硬化(AS)是一种慢性血管炎性疾病,由内皮细胞损伤激活引起,随后出现脂质代谢异常形成脂质积累、内皮功能障碍,血小板活化、血管炎症反应、钙化和纤维化,触发血管狭窄和炎症途径的激活.补体系统是先天免疫的重要组成部分,补体C1q在AS中发挥着双重作用,在经典途径中C1q激活补体系统促进AS的发展,然而C1q还可以通过减少泡沫细胞的形成、减少巨噬细胞的促炎细胞因子信号和增强巨噬细胞的存活能力等多种机制抑制AS的发展.并且C1q作为动脉粥样硬化的新刺激风险基因,利用C1q相关基因的分子特征开发基因标记,可能有效的诊断和预测AS的发生和进展.本文就补体C1q在AS中发挥的作用及其作为预测AS的基因进行简要综述.
Abstract
Atherosclerosis(AS)is a chronic vasculitic disease caused by injury and activation of endothelial cells,followed by abnormal lipid metabolism resulting in lipid accumulation,endothelial dysfunction,platelet activation,vascular inflammatory response,calcification and fibrosis,triggering vascular stenosis and activation of inflammatory pathways.The complement system is an important part of innate immunity,and complement C1q plays a dual role in AS.In the classical pathway,C1q activates the complement system to promote the development of AS.However,C1q can also inhibit the development of AS through various mechanisms such as reducing the formation of foam cells,reducing the pro-inflammatory cytokine signal of macrophages and enhancing the survival ability of macrophages.In addition,as a new stimulating risk gene for atherosclerosis,the development of genetic markers based on the molecular characteristics of C1q-related genes may effectively diagnose and predict the occurrence and progression of AS.This article reviews the role of complement C1q in AS and its role AS a gene for predicting AS.
