Effect of adipose mesenchymal stem cells on K562 cell proliferation
Objective To establish a co-culture system of adipose-derived stem cells(ADSCs)and K562 cells,and to explore the effect of adipose mesenchymal stem cells on the proliferation of K562 cells.Methods Primary isolation of ADSCs was cultured to the 3rd generation and identified.ADSCs and K562 cells were co-cultured,K562 cell wells were used as control alone,and 24 wells were seeded respectively.Six-well cells were selected on days the 1st,3rd,5th,and 7th,and cells were counted and cell growth curves were plotted.The generalized estimation equation was used to statistically ana-lyze the repeated measurement data.Results ADSCs were isolated and successfully identified.The cell growth curve showed that with the culture time from the 0th day to the 7th day,the proliferation rate of K562 cells in the single culture group and the co-culture group was getting faster and faster,and the average number of K562 cells in the single culture group was(2.85±0.08)×104/mL,(10.70±0.36)×104/mL,and(39.00±0.30)× 104/mL,the average number of K562 cells in the co-cul-ture group were(2.67±0.11)×104/mL,(10.01±0.19)×104/mL,and(38.00±0.37)× 104/mL,respectively,and the differences in the number of proliferating cells between the two groups on the same day were statistically significant(all P<0.001).Conclu-sion ADSCs have an inhibitory effect on the proliferation of K562 cells.
